The similarity of anatomy and physiological structures between Taihu pig andhuman makes it be an ideal organism as model of human disease. The applications ofTaihu pig in medical research are more and more. This study selected on local goodvariety,that is Taihu,analyzed genetic structure and physiological profiling,then builtExperimental Taihu pig strain. Made the ventricular fibrillation model to evaluate andstudy the defibrillation electrodes.This study analyzed the genetic background of closed colony Taihu pig and Binaryhybridization Taihu pig, which used microsatellite molecular marker.Selected14pair ofprimers and amplified microsatellite loci of Taihu pig and Binary hybridization Taihupig. The number of alleles respectively were1-5and2-5,the effective number of allelesvariation range respectively were1.0000~3.9320and1.1245~4.4806, the averageeffective number of alleles respectively were2.2805and2.1936, the average observedheterozygosity respectively were0.6176and0.5540, the average Shannon’s Informationindex were0.8217and0.8750, the average expected heterozygosity respectively were0.4847and0.4927.The observed heterozygosity and effective number of alleles of Taihupig were higher, conjecturing Taihu pig’s recessive allele was lost during hybrid process,while Binary hybridization Taihu pig’s heritable variation degree was higher.The polymorphism information content variation range of Taihu pig and Binaryhybridization Taihu pig respectively were1.0000~0.7047and0.0554~0.7403. While theaverage polymorphism information content variation range of Taihu pig and Binaryhybridization Taihu pig respectively were0.4362and0.4415.The polymorphisminformation content of Binary hybridization Taihu pig was higher, both were middlepolymorphism. Genetic diversity of Taihu pig is ample, genetic system is stable so thatbe not susceptible affected by environment. Taihu pig accords with the geneticcharacters of closed colony. Taihu pig at different age between2months、4months and6months was measuredthe blood physiological indexes. Compared to other month, there was significantdifference at2month of age Taihu pig’s platelet distribution width and neutrophilratio,indicating that low platelet level had an effect on coagulation function. So thatIt’s better to raise the ratio of high protein fodder. The WBC、RBC was low to someextent, conjecturing the main reason was the difference on geographic location andclimate pattern, though were both in the normal range. Analyzed the indexes on atdifferent gender but same months of age, it showed no significant difference.Cardiac catheter was used to induce and build ventricular fibrillation model onthree different weight groups of Taihu pigs. Kept P-500series electrodes as control,utilized electrodes of P-700and P-500to do external defibrillate and resuscitation invitro separately. Counted the rate of successful defibrillation, Did Doppler ultrasoundmeasurement to evaluate the animals’ myocardial function before and after defibrillation,did the gross anatomy and pathological examination after experiments. Between thecontrol groups and experimental groups, It showed no obvious significance differenceon the rate of successful defibrillation. Among the three groups, there was no obvioussignificance difference prior to and after experiment (P>0.05) on left ventricularejection fraction(EF)、fraction area change(FAC) and left ventricular end-systolicthickness(LVPWS). No obvious abnormal appearance was found in gross anatomy andpathological examination. Taihu pig can effectively assess the function and side effectof electrodes. There is also some different response among individuals, according withthe character of closed colony. Taihu pig is appropriate to be important experimentalanimal on cardiovascular research.
