Objective: To explore the expression of MMP-1, MMP-2, endotoxin and CD14inthe biliary atresia liver tissue in different levels, and provide a theoretical basis oftreatment to children with biliary atresia liver fibrosis in the level of molecular biology.Methods: The experimental group has17cases of children with biliary atresialiver fibrosis, according to the Ohkuma’s grading standards, grade I has6cases, grade IIhas4cases, grade III has5cases and grade IV has2cases. The negative group has7cases. Detect the protein expression and tissue localization of MMP-1, MMP-2,endotoxin and CD14by ELISA and immunohistochemistry method (S-P). Use the SAS8.0statistical software to analyze.Results: To detect the protein expression of MMP-1, MMP-2, endotoxin andCD14of17patients with biliary atresia liver tissues of the experimental group and7patients with negative control group, the results show: I, II and III levels in MMP-2protein are different with the control group (3724±1205,3427±867,2819±824Vs2318±528ng/ml)(P <0.05);The presences of I, II and III in CD14protein aredifferent with the control group (4696±1652,4308±1428,3879±1453Vs2634±612ng/ml)(P <0.05). Four factors in the expression amount of Class IV with the controlgroup are no differences in MMP-1protein (2723±845Vs2703±871ng/ml),MMP-2protein (2467±613Vs2318±528ng/ml), endotoxin (6.2±4.1Vs6.16±4.81EU/ml) and CD14protein (2722±767Vs2634±612ng/ml)(P>0.05). Fromthe BA liver fibrosis of view, from Class I to Class IV, MMP-2and CD14proteinexpression was a gradual downward trend, while the expression of MMP-1protein andendotoxin in the experimental group and negative control group was no difference.MMP-1, MMP-2, endotoxin and CD14protein expression detected by immuno-histochemistry biliary atresia liver tissue of17cases of the experimental group, theresults show: MMP-1and MMP-2expression in the portal area fibrosis with theendotoxin and CD14protein expression in pseudolobules and promote liver fibrosis. Conclusion:1. From Class I to Class IV of the BA liver fibrosis of view, MMP-2and CD14protein expression was a gradual downward trend, while the expression ofMMP-1protein and endotoxin in the experimental group and negative control groupwas no difference.2. MMP-2and CD14regulated the formation of the BA liver fibrosis negativly,MMP-1and endotoxin were not associated with the formation of the BA liver fibrosis.
