| objective:To investigate the proliferation and apoptotic effects of parathyroid hormone (PTH) and Anti-Parathyroid Hormone Receptorl (anti-PTHR1) antibody on the human medullary Thyroid cancer cell line TT and its changes, including the changes of cellular morphosis, cell proliferation rate. early apoptotic rate, and PTHR mRNA expression, and to provide further evidence for the clinical application of PTH and anti-PTHRl antibody on MTC.Methods:human medullary Thyroid carcinoma TT cell line was cultured and treatment with different concentrations of PTH and anti-PTHRl antibody in different groups. observed respectively under light and electron microscope. inhibitory rate of cell proliferation was detected with MTT assay, Apoptotic rate were detected with DAPI staining. Early apoptotic rate was detected by flow cytometry (FCM). And PTHR mRNA expression was detected by RT-PCR.Results:1) Typical morphologic changes of apoptotic cells were observed under light and electronic microscope in treatment group.2) proliferation rate detected with MTT assay in treatment group was lower than that in control group (p<0.05).3) apoptotic rate in treatment group was higher than that in control group (P<0.05).4) Changes intracellular PTHR expression level detected by RT-PCR in treatment group were observed (P<0.05).5) PTHR expression level in PTH treatment group was increased obviously, with the decrease of anti-PTHR1antibody treatment group by RT-PCR detection.Conclusion:2.0μmol/L dose of PTH and1.0μmol/L dose of anti-PTHR1antibody can inhibit the proliferation of TT cells, block cell cycle, significantly decrease proliferation cell exponent, It plays an important role in inducing apoptosis. The expression changes of PTHR may be one of the mechanism of human medullary carcinoma cell line TTs apoptosis. |
