Objective To investigate the protective effect of dexmedetomidine againstischemia-reperfusion(IR)injury in mice liver and its mechanism.Methods Forty C57BL/6mice were randomly divided into sham-operation group (SO),sham-operation anddexmedetomidine group (SO+Dex),ischemia reperfusion group(IR),ischemia reperfusionand dexmedetomidine group(IR+Dex). The mice in SO and SO+Dex group wereabdominal laparotomy off after60min.The mice in IR and IR+Dex group received a60min occlusion of70%hepatic blood inflow,followed by6h reperfusion.The mice in IRand IR+Dex group were intraperitoneally injected30min before surgery withdexmedetomidine given25ug/kg. The mice in SO and IR group were intraperitoneallyinjected30min before surgery with equal volume of saline. Serum activities of alanineaminotransferase(ALT) and aspartate aminotransferase(AST) were determined by ratemethod.Serum concentrations of tumor necrosis factor-α(TNF-α) and interleukin-6(IL-6)were detected by enzyme linked immunosorbent assay (ELISA).The liver histologicalchanges were examined after hematoxylin-eosin (HE) staining and the liver cell apoptosiswere examined by terminal dexynucleotidyl transferase-mediated dUTP nick end labeling.Results Serum levels of ALT and AST,serum concentrations of IL-6and TNF-α weremarkedly decreased (P<0.05) in IR+Dex group compared with the IR group.The injury ofhepatocytes was ameliorated in IR+Dex group compared with the IR group. The hepatocyteapoptosis index was significantly lower(P<0.01) in IR+Dex group compared with the IRgroup.The indicators were not statistically different between SO group SO+Dexgroup.Conclusion Dexmedetomidine can reduce the ischemia-reperfusion injury of miceliver by inhibiting the expression of proinflammation eytokines....
