Objective:To explore the relationship between Men1and menin expression and tumor types, wedesign the project to study the expression levels and subcellular localization of Men1andmenin in thirteen types of non-endocrine tumor cells. We try to find the key regulatorymolecules of Men1and menin expression in non-endocrine tumor cells, and lay thefoundation for development of novel strategies of non-endocrine tumor treatment andprevention in the future.Methods:RT-PCR and Western blot were performed to detect the expression level of Men1mRNA and menin in thirteen types of non-endocrine tumor cells, including AGS,BGC-823, SGC-7901and MNK-28(gastric cancer), GES-1(transformed gastricepithelium), Huh7and Hep3B2.1-7(liver cancer), MDA-MB-231and MCF-7(breastcancer), SW480(colon cancer), SKOV-3(ovarian cancer), HeLa (cervical cancer) andSGH44(brain glioma) cells. Western blot and Immunofluorescence were performed todetect the location of menin in thirteen types of non-endocrine tumor cells. ChooseMKN-28and GES-1with octreotide Treatment, then to detect the expression level and thelocation of Men1mRNA and menin in the two cells with Western blot andImmunofluorescence.Result:Men1mRNA and menin was positively expressed in13human cancer cell linesexamined in this experiment, especially in the gastric cancer cell lines. The expression ofmenin mRNA was gradually increased from AGS, BGC-823, SGC-7901and MNK-28to GES-1.Menin was located primarily in the nucleus of all cell lines examined in thisexperiment. However, in the four human cancer cell lines (GES-1, MCF-7, SGH44andHeLa), menin was localized not only in the nucleus, but also in the cytosol and membrane.The relative protein expression levels of menin in the GES-1and MKN-28cell lines weresignifcantly increased in the octreotide group compared with the control group. Moreover,more menin protein was located in the cytosol with octreotide treatment.Conclusion:1、Men1mRNA and menin were expressed in all the thirteen types of non-endocrinetumor cells, and the expression is related to the differentiation degree.2、Menin was located primarily in the nucleus of the thirteen types of non-endocrinetumor cells. However, in the four human cancer cell lines (GES-1, MCF-7, SGH44andHeLa), menin was localized not only in the nucleus, but also in the cytosol and membrane.3、Octreotide promote the expression of menin in tumor cells.
