OBJECTIVETo explore the effect of Bone Marrow Mesenchymal Stem Cell and CD133+renal cell in Acute Renal Injury induced by Ischemia reperfusion.METHODS1. C57BL/6mouse marrow mesenchymal stem cells(MSCs) was separated by percoll density gradient centrifugation and adherent separation. CD133+cells were separated in the kidney by immunomagnetic selection and adherent separation.2.40male C57BL/6mice were divided into normal control group, group I/R (clipping bilateral renal pedicle and then reopening after10minutes), group I/R+MSCs (clipping bilateral renal pedicle and then reopening after10minutes,at the same time immediately injected MSCs into mice through tail vein), I/R+CD133+renal cell group (clipping bilateral renal pedicle and then reopening after10min,at the same time immediately injected CD133+cells into mice through tail vein),10rats in each group.2mice in each group were sacrificed on first days, second days, third days, seventh days, fourteenth days after the mice model established.3. The arterial blood of the mice was detected for the blood urea nitrogen (BUN) and creatinine (Cr) levels. The renal tissue were prepared for kidney tissue slices and HE staining, observed the change of renal pathological, and evaluated kidney tissue acute tubular necrosis (ATN score) of kidney tissue.The levels of tumor necrosis factor-a(TNF-a) and hepatocyte growth factor (HGF) was detected by enzyme-linked immune sorbent assay(ELISA). RESULTS:1. The level of the BUN, Cr, the tumor necrosis factor-α (TNF-α) and the score of ATN of I/R+MSCs group were higher than those of the normal group, but it was lower than those in I/R group (on the seventh days after operation was the most significantly (P<0.05)). The level of the hepatocyte growth factor (HGF) of I/R+MSCs group were lower than those of the normal group, but it was higher than those in I/R group (on the seventh days after operation was the most significantly (P<0.05)).2. The level of the BUN, Cr, the tumor necrosis factor-α (TNF-α) and the score of ATN of I/R+CD133+renal cell group were higher than those of the normal group, but it was lower than those in I/R group (on the third days after operation was the most significantly). The level of the hepatocyte growth factor (HGF) of I/R+CD133+renal cell group were lower than those of the normal group, but it was higher than those in I/R group (on the third days after operation was the most significantly (P<O.05)).3. The level of the BUN, Cr, the tumor necrosis factor-a (TNF-a) and the score of ATN in I/R+CD133+renal cell group were lower than that were in I/R+MSCs group.The level of the hepatocyte growth factor (HGF) in I/R+CD133+renal cell group were higher than that was in I/R+MSCs group (P<0.05).CONCLUSIONS:Exogenous bone marrow mesenchymal stem cells (MSCs) and CD133+renal cells may regulate cytokines secretion and improve microenvironment in injury renal by endocrine which can contribute to the recovery of acute renal injury, while the CD133+renal cells may be more effective than MSCs.
