The thymus is a specialized primary lymphoid organ in the mouse, generating mature T lymphocytes to the peripheral circulation, it sustains autoimmune stability. According to the theory of central tolerance, peripheral tolerance on allogeneic antigens can be induced when T cells are exposed to the same antigen during development in the thymus. At the same time, blood-thymus barrier proved a stable microenvironment for T cell development, foreign macromolecular substances and antigens can not entry the thymus easily, so the immune mechanism of the thymus was not attentioned before. In orader to make a better study of the thymic defence function to the exogenous antigen,we used the motheods of minimally invasive injection and staining of tissue sections and so on, to explore the immune response of mice thymus of nanoscale xenobiotics and xenogeneic cells occur. The main conclusions are as follows:(1)Intrathymic injection of nanometer material is first concentrated in the medulla of thymus, and then gradually transferred to the interlobular areas; in the thymus of nanometer materials had no observable effect on lymphocyte, did not induce immune response occurred as injection of xenogeneic cells that can be induced to swallow, but on cell phagocytosis and transfer, and very difficult degradation in cells.(2) tumor block formed by the injection of tumor cells in the thymus, mainly occurs in the cortex, in6-13days after injection can induce lymphocyte infiltration in tumor block, resulting in tumor necrosis.(3) the immune tolerance in the thymus of antigens of previous research interpretation, has become a classic theory, but we found that immune in xenogeneic antigen mediated specificity in the thymus of exclusion, further exploration of this mechanism, will become a core problem of the research of immunology. It has important scientific significance in the viral infections, autoimmune diseases, tumor immunity and transplantation immunity.
