Objectives: To study hypoglycemic effect on the type II diabeticmice,and Protective effect on the type II diabetic micehypolipidemic effect and diabetic nephropathy treated by Momordicasaponins and berberine mixture;To explore its mechanism from theaspects of glucose metabolism, lipid metabolism, and renalfunction,and provide a theoretical basis for the clinicalprevention about complications of diabetes by the application ofMomordica saponins and Berberine mixture.Method: Continuous feeding with high fat diet (diet78.8%, eggyolk powder10%, lard10%,1%cholesterol, cholate0.2%) for2weeksto establish hyperlipidemia model mice of Kunming species. Througha single high dose (200mg/kg) by intraperitoneal injection ofstreptozotocin (STZ) model to replicate the experimental type2diabetes mice. Diabetic model mice were randomly assigned to thecontrol model group, rosiglitazone the positive control group,Momordica saponins group, Momordica saponins and berberine mixturegroup, and the establishment of the blank control group. Continuousafter12weeks, determination of mice weight, liver weight, kidney weight. All provisions of kit and strict application of biochemicalanalysis method do the detection of blood glucose concentration(GLU), glucose tolerance, total cholesterol (TC), triglyceride(TG), serum creatinine (SCr), creatinine (UCr) and other indicators.And in accordance with the provisions of ELISA Kit ELISA., strictapplication of double antibody sandwich method of one step do thedetection of transforming growth factor-β1(TGF-β1) and urinarymicroalbumin (mALB), glycosylated hemoglobin (GHbAlc) and otherindicators.Results:1There was96experimental Ⅱ diabetes mouse model to copy thesuccess among100build model miceAfter4weeks of high-fat feeding and fasting12h, chain ureaon building model mice with cephalosporins by intraperitonealinjection of sodium citrate, citric acid solution, injection chainurea with cephalosporins in3days, fasting12h, the tail vein bloodtest fasting blood sugar, Building100mice,96fasting plasmaglucose or greater tendency for11.1/L.2Good general condition were observed in the group of Momordicasaponin and berberine mixtureFrom the point of the observation of general condition, a mixtureof Momordica saponin and berberine can make experimental Ⅱdiabetes mice spirit better, increase exercise, weight gain. Afterbuilding success, in addition to the normal control group (groupA) of the rest of the each weight of mice has obvious downward trend,and it was extremely significant difference (P <0.01) compared withthe normal control group (group A). For3weeks, rosiglitazone group(group C), Momordica saponin group (group D), the mixture of Momordica saponin and berberine group (group E) mice body weighthad a differently degree of rebound, there were no significantdifference (P>0.05) compared with model control group (group B).For6weeks and9weeks,12weeks, rosiglitazone group (group C),Momordica saponin group (group D), the mixture of Momordica saponinand berberine group (group E) mice weight rebounded significantly,there were significant difference (P <0.05)compared with modelcontrol group (group B).3Blood sugar reducing effect was obvioused in the group ofMomordica saponin and berberine mixtureType Ⅱ diabetic mouse model to replicate the success, comparedwith the normal control group, model group mice fasting bloodglucose values were significantly elevated (P<0.01). After theAdministration2weeks, the positive control group, group ofMomordica saponins, saponins and berberine mixture group comparedwith model group, each group of mice fasting blood glucose valuehad declined in varying degrees. After4weeks, compared with themodel group, the positive control group, group of Momordicasaponins, saponins and berberine mixture group mice fasting bloodglucose values had decreased significantly (P<0.01). From theAdministration for4weeks, until12weeks of administration, inaddition to blood glucose of model group mice still has little scopeto rise, other groups mice blood glucose were the basic value rangeremained stable in blood glucose. Among them, the obvious effectwas the group of Momordica saponins and berberine mixture.4The group of Momordica saponin and berberine mixturesignificantly improved the glucose tolerance of diabetic miceAccording by the glucose tolerance test, the mice in the normal control group0.5h postprandial blood glucose value reached itspeak, postprandial1.5h blood glucose values had basically returnedto the level before the meal. Model group mice reached peakpostprandial blood glucose values in1h, it had been in a state ofhigh blood sugar. Momordica saponins mice in the postprandial0.5hblood glucose value reached its peak, but in the postprandial1hblood glucose decreased not obviously,2h postprandial bloodglucose returned to level before meal. Positive control mice,Momordica saponins and berberine mixture group mice reached peakpostprandial blood glucose values in0.5h, basically the same asin1h when the two groups of mice postprandial blood glucose levels,in the postprandial2h when the two groups of mice blood glucoselevels returned to guide before meal, blood glucose values werehigher than the blank control group.5Momordica saponin and berberine mixture can significantlyreduce the level of total cholesterol and triglyceride fromdiabetes miceBy4weeks of drug treatment, each group of mice compared withthe model group, the positive control group, group of Momordicasaponins, Momordica saponins and berberine mixture group cansignificantly reduce the level of TC and TG in serum of diabetesmice(P<0.01)6Momordica saponin and berberine mixture can significantlyreduce the levels of transforming growth factor beta1(TGF-β1),urine micro protein(mALB) and glycated hemoglobin (GHbAlc)Through12weeks of drug treatment, each group comparing withmodel control group, positive control group, Momordica saponinsgroup, Momordica saponins and berberine mixture group mice transforming growth factor β1(TGF-β1), microalbuminuria(mALB), glycosylated hemoglobin (GHbAlc) decreased significantly(P<0.01). Among them, the most significant mixture of Momordicasaponins and berberine effect.7The diabetes mice serum creatinine (SCr), urine creatinine (UCr)was remarkable effect treated by Momordica saponins and berberinemixtureThrough12weeks of drug treatment, each group comparing withmodel control group, positive control group, group of Momordicasaponins, saponins and berberine mixture group mice serumcreatinine (SCr) were significantly decreased (P<0.01), creatinine(UCr) levels were significantly increased (P<0.01). Among them,the most significant effect was the group of Momordica saponinsand berberine mixture.8Diabetes renal protection effect was obvious in the group ofMomordica saponin and berberine mixtureBy12weeks of medication, observing the normal control group(group A), model control group (group B), the mixture of Momordicasaponin and berberine group (group E) in mice kidney biopsy, foundthat the normal control group (group A) glomerular, renal tubularparts not seen obvious abnormity; Model control group (group B)increase the bowman’s capsule cavity, a significant deformation;Severe renal tubular injury, inflammatory swelling of epithelialcells, malpighian tube have protein; Momordica saponin and mixtureof berberine group (group E) inflammation of the renal tubular epithelial cells recovered, but some of the malpighian tube and asmall amount of protein.Conclusion: The mixture of Momordica saponins and berberine couldsignificantly regulate the experimental diabetic mice in glucosemetabolism, lipid metabolism and pretection renal function, themixture of Momordica saponins and berberine had obvious effect onthe type Ⅱ diabetic mice hypoglycemic effect and prevention ofdiabetic complications.
