Objective: Backusella circina belong to the Eumycota, zygomycetessubphylum, Mucorales, widely distributed in the nature. Mucorales fungus ofthe zygomycetes is a kind of important in the economic benefit, can be dividedinto14families,55genera and more than400, the vast majority of themucorales distribution in the soil organic matter, and the animal corpse. In thehuman pathogenic aspects,general for condition pathogenic bacteria,when thehuman body in serious illness or late malignant disease, the body resistancefall into human body, cause human body shallow and deep fungal disease,domestic and foreign related mucor infection reported in recent yearsgradually increased, but the strain temporarily no related reports in theliterature.This research use strain separation self court1cases of leukemiapatients infected with the face of black scab, by the Chinese academy ofsciences institute of microorganisms identified preliminarily as Backusellacircina.On the one hand,through the analysis of some simple physiologyexperiment to understand the basic characteristics of the bacteria; On the otherhand,through the Backusella circina in five different medium morphologywere observed, to find more suitable for the growth of environmentaltemperature and medium.Methods:1Colonial morphology observation1.1Five kinds of medium colony morphology observation To save strain ofour hospital dermatological department under normal temperature recoveryforty-eight hours.Put the isolation under the ordinary temperature rewarmabout forty-eight hours and inoculate it respectively on the media ofSabouraud′s Agar﹙SDA﹚,Malt Extract Agar﹙MEA﹚,Patato Glucose Agar﹙PDA﹚,Corn Meal Agar﹙CMA﹚and Czapek’s Agar﹙CZA﹚,andculture at27℃、32℃、37℃and40℃for sixty hours, then observe itsdevelopment and measure the diameters of colnies at12hours、36hours、60hours.1.2Slide culture Inoculate the strain on the microculture of the above fivekinds of media at27℃and37℃for sixty hours and observe the growth ofhypha and spores at60hours.1.3Observation under light microscope Put some samples taken from thefive kinds of media colonies on the glass slide, cover them with slip whendissolved in the solution of cotton blue stain, then observe it under lightmicroscope.1.4Observation under scanning electron microscope The samples takenfrom the medium of SDA in the27℃were fixed in the solution of4%glutaric dialdehyde and prepared through routine procedure and examinedunder scanning electron microscope.2Physiology experiments2.1Urease test Inoculate the strain on the Urea agar,and culture it at theordinary temperature for four days,then observe the change of the mediumcolor.Results:1Five kinds of medium strain general morphology observation of fivekinds of medium in four kinds of temperature all have colony growth, colonyare fluffy white wool shape. Strain in the SDA, MEA, PDA colony in fourdifferent temperature, colony growth speed are faster than CZA and CMA ofthe growth rate in the same temperature.2In the five kinds of medium colony diameter Backusella circina infive kinds of medium all have growth, growth rate and is proportional to thetime. colonies under the same temperature in a different medium diametercompared significant difference (P <0.05)(Table1), the diameter of eachmedium are expressed as mean and standard deviation (Table2,3,4), the SDAgrowth fast.27℃and32℃in the same medium colony diameter without significantly with sex differences (P>0.05)(Table6),27℃and32℃,respectively, and37℃,40℃relative comparison, there is a significantdifference (P <0.05)(Table6),40℃under colonies almost no growth (notmeasure the growth diameter).3Morphology under light microscope It showed abundant separationbranched hyphae. The mycelium were of rather diverse structure, with clampconnections and curved lateral pegs. It showed the rhizoid formation in thehyphae end, Visible single or gather piles size circular spore.4Morphology under scanning electron microscopy Visible rich branchwithout separation hypha, wider, structural differences, degree of finish is notuniform, the diameter is about2~6um, circular sporangia peripheral isradiation prod wart structure, wart between no connection, spore assumes thecircular, the diameter is about1.5~3um.5Physiology experiments The test of urease reaction was positive.Conclusion:1Backusella circina colonies only one form at each temperature, as awhite wool-like, grown on a sand castle’s medium, potato dextrose agarmedium, Malt agar culture condition, and the sand castle’s mediumthe fastestgrowing, and27℃,32℃, the growth rate is better than37℃,40℃.2Urease agar medium positive test results.
