Cardiac carcinoma is one of the most common malignant tumors in theworld. And it is regarded as a special type of gastric cancer, neither distalesophageal carcinoma nor ordinary gastric carcinoma. Its characters dependon its unique anatomical structure, while the onset of special parts and specialbiological behavior determines its diagnosis, surgical procedure, and poorprognosis.China is a high incidence region of cardiac carcinoma. Its mortality ratehas reached the second place, but so far, the cause of this kind of malignanttumor is still unclear. In the past, scholars considered it having related to theenvironment, diet, genetic, chronic atrophic gastritis and helicobacter pylorifactors. However many studies have showed that the biological basis of thedisease is closely related to the tumor stem cells (TSC) recently, scholars tendto think it as a kind of stem cell disease, so the research of the TSC and thespecific antigen is more and more important.CD24and ABCG2have been widely accepted by more and morescholars as specific markers of TSC. CD24is a highly glycosylation moleculeconsisting with27different amino acids. Lots of researches showed that CD24was related with many malignant tumors, including breast carcinoma, ovariancarcinoma, esophageal carcinoma, gastric carcinoma and colorectal carcinomaetc, especially in digestive tumors. It regulates the cell proliferation anddifferentiation in tumors. ABCG2protein(ATP-binding cassette super familyof G member2)is a member of ABC (ATP-binding cassette) super familyprotein, involved in multi-drug resistance of tumors, it is one of the factorswhich induce the bad effect of chemotherapy in patients. So it has importantsignificance to explore its relationship with the clinical pathological features.Few reports were found about CD24and ABCG2in cardiac adenocarcin- oma. In this study, we explore the expression of CD24and ABCG2in cardiacadenocarcinoma by immunohistochemistry and RT-PCR technology, andexplain the essential role of TSC in cardiac adenocarcinoma.Objective: To investigate the expression of ABCG2and CD24in cardiacadenocarcinoma, explore the relationship between the two molecules andother clinicopathological features, analyze the interact between CD24andABCG2.Methods:1.The expressions of CD24and ABCG2in140cases ofcardiac adenocarcinoma were detected by using immunohistochemistry (spmethod). All the cases were divided into different groups according to degreeof differentiation, age, gender, Clinical staging and pathological morphology.After getting the positive rate of two molecules in each groups, we analyzedthe relationship between two molecules and clinicopathological features, andfuther explored the essential role between the two molecules and cardiacadenocarcinoma biological behavior.2. Total RNAs were extracted from26cases of fresh carcinoma tissues and adjacent mucosa, each groups weredivided by different clinical staging, different degree of differentiation. Theexpression of CD24and ABCG2were detected by using RT-PCR and theresults were shown as relative quantity. We analyzed the invasion of CD24and ABCG2by comparing the difference in each groups.Results:1Immunohistochemical results:1.1The expression of CD24and ABCG2was significantly higher incarcinoma tissues than that in adjacent mucosa (P<0.05), and thoseexpressions were associated with tumor differentiation (P<0.05). CD24wasalso closely related with the characters of carcinoma invasion, lymph nodemetastasis, clinical stages and nerves invasion (P<0.05), while no relation withage, gender, vascular blots and residual situation. ABCG2was not related withany other clinicopathological features except differentiation (P>0.05).1.2The expression of CD24in two pathological types, Lauren: Diffuse typewas higher than intestinal type; WHO: The expressions of CD24were different in tubular adenocarcinoma, polypoid adenocarcinoma, mucinousadenocarcinoma and low adhesion adenocarcinoma (P<0.05).1.3The expression of ABCG2in two pathological types, Lauren: Diffuse typewas higher than intestinal type; WHO: The expressions of the ABCG2weredifferent in tubular adenocarcinoma, polypoid adenocarcinoma, mucinousadenocarcinoma and low adhesion adenocarcinoma (P<0.05).1.4CD24had no significant relation with ABCG2(P>0.05).2RT-PCR results demonstrated that the expressions of CD24and ABCG2were markedly higher in carcinoma tissues than those in adjacent mucosa (P <0.05).The expressions of CD24and ABCG2in poorly differentiatedadenocarcinoma were significantly higher than those in medium/highdifferentiated adenocarcinoma (P<0.05).The relative quantity from Ⅰstagingto Ⅳ staging was a raised procession (P<0.05). ABCG2was independent withTNM staging (P>0.05).Conclusions: The results that CD24is closely related with the charactersof tumor differentiation, carcinoma invasion, lymph node metastasis andclinical staging demonstrate that CD24may be involved in cardiacadenocarcinoma malignant process, the prognosis of patients with the higherexpress may be worse. The expression of CD24is related with pathologicalmorphology. ABCG2is closely related with tumor differentiation andpathological morphology.
