Since1899Liverpool Prison William Sullivan doctors found a higher rate of alcoholism pregnant women gave birth to birth defects, to the University of Washington in1973, Dr. Kenneth Lyons Jones and David W. Smith formally proposed the "fetal alcohol syndrome".There is growing recognition of the impact of alcohol on fetal development, especially for fetal brain damage. Sphingomyelin synthase2gene knockout (sphingomyelin synthase2knockout, SMS2(?)) mice to establish prenatal alcohol exposure model by observing the exposure of alcohol during pregnancy, the SMS2gene moss cells lost among the relationship to reveal the target of alcohol nerve toxicological effects, and further explore the moss physiological function of cells and its relevance. In addition, through the study of the model to help people better understanding of alcohol in pregnancy offspring central nervous system damage caused by abuse, but also for the future treatment and prevention of alcoholic dementia and alcohol epilepsy and other neurological diseases, new treatment ideas treatment strategies and appropriate treatment.Objective:The study of alcohol during pregnancy exposed mice dentate gyrus mossy cells and camparing on the function of SMS2(?) role in the process to do a more in-depth observation. Methods:PCR method was used to identify each packet pups using SMS2(?) mice to establish pregnancy alcohol exposure model, to collect P14pups, water maze experiments, observe the time of finding platform pups; then using immunofluorescence staining each packetpups dentate gyrus mossy cells changes in the number of the immunoblotting P14hippocampus organizations GluR2/3relative expression level of the protein.Results:1.Alcohol group compared with the control group, the offspring search time was significantly longer (P<0.05). Knockout pups find time and wild group there was no significant difference (P>0.05).2.Alcohol pups GluR2/3the number of positive cells was significantly less than the control group (P<0.05), and SMS2(?) group and WT pups compared SMS2gene knockout pups GluR2/3positive cells was no significant difference (P>0.05).Conclusion:1.Alcohol exposure causes the moss cell loss, moss cells reduce loss of recent memory;2.Alcohol exposure can reduce the relative expression levels of the GluR2/3;3.SMS2(?) the moss cell loss and GluR2/3relative expression levels decreased relatively minor role.
