BACKGROUND:Type2diabetes mellitus (type2diabetic mellitus, T2DM) is a metabolic disorders and can cause multi-system damage with the progression of the disease,has now become a global epidemic disease that endanger human health.And obesity is a major predictor of the risk of type2diabetes in the future, involved in a variety of metabolic diseases,including type2diabetes,insulin resistance,dyslipidemia and hypertension,but the molecular mechanism is unclear.Fat mass and obesity-associated gene FTO is a fat candidate genes analysed through a genome-wide association,by acting on BMI associated with type2diabetes. But there were also study reported that FTO gene and type2diabetes had strong independent correlation,and this correlation was also confirmed in Asian populations,but the results were not very consistent. Yajnik found that The indo-european race’s FTO rs9939609allele A was associated with type2diabetes independence of BMI,Chang reported FTO gene rs9939609polymorphism had correlation with obesity and BMI in China Taiwan adults; Tran Quang Binh found FTO gene rs9939609increased risk of type2diabetes in Vietnam person, and the correlation was independent of BMI.Because of in recent years,most of the researchs was to investigate the relationship between the FTO gene loci polymorphism with obesity, and the results of all the study were not the same,so the purpose of this experiment is to observe the FTO gene expression level in patients with type2diabetes,And in order to provide more theoretical basis of formation mechanism and prevention method of type2diabetes.OBJECTIVE:Measuring the expression of peripheral blood lymphocytes FTO mRNA and serum FTO protein in patients with type2diabetes,explore FTO gene expression levels and the correlation of type2diabetes.METHODS:135patients with newly diagnosed type2diabetes patients in Henan Province is selected and divided into obese group (60cases) and non-obese group (75cases),our hospital medical diagnosis of normal persons as normal control group (51cases),the application of reverse transcription-polymerase chain reaction (RT-PCR) method for determining FTO mRNA expression,enzyme-linked immunosorbent assay (ELISA) determination of FTO protein expression levels,gel electrophoresis scanning using computer analysis software with the result of electrophoresis analysis of gray value contrast,the above experimental results using SPSS17.0software for statistical analysis.RESULTS:1.compared with normal control group,the obese T2DM group of body weight,BMI,waist circumference,waist-hip ratio,diastolic pressure,FPG,FINS,HOMA IR,OGTT2h,HbA1C,24halb,TG,TC,LDL-C increased(P<0.05), HDL-C decreased (P<0.05);T2DM non-obese group of diastolic blood pressure,FINS,HOMA IR,FPG,OGTT2h,HbA1C,24halb,TG,TC,LDL-C increased(P<0.05),HDL-C reduce, hs-CRP (P<0.05). comparison between T2DM two groups:the obese T2DM group of body weight, BMI, waist circumference, waist-hip ratio, OGTT2h, TG,FINS, HOMA IR is higher than the non-obese group(P<0.05).2.compared with normal control group, The amount of FTO mRNA and protein expression are increased(P<0.05) in type2diabetes with obese and non-obese group.3.The expression level of is positively correlated with systolic pressure,diastolic pressure,FPG,FINS,HOMA-IR,24halb,OGTT2h,HbAlC,and TC(P<0.05),and negatively correlated with HDL-L(P<0.05).4. Logistic regression analysis showed:BMI, TG, HOMA-IR, the expression levels of FTO gene and FTO protein were the major risk factors for type2diabetes.CONCLUSION:the FTO expression levels of type2diabetes is significantly increased, FTO gene may be independent of BMI affect insulin secretion and insulin resistance, may be one of early predictors of type2diabetes.
