Objective To discuss H1receptor blocker acting as prophylactic against adverse reactions of protamine about the effect of prophylactic administration and changes of serum amounts of C3、C4after extracorporeal circulation. Methods To divide80patients into control group(A) and prophylactic group(B). Group A without receiving H1receptor blocker. Patients in group B were prophylactically given intravenous H1receptor blocker0.2mg-kg-1diphenhydramine. Serum levels of C3、C4were measured and hemodynamic paramelars were recorded before and after protamine(AP). Results After AP, the SBP, DBP, MAP and SVR decreased significantly (P<0.01) in group A, but with no abvious changes in group B(P>0.05). The differences were remarkable between the two groups(P<0.01). Serum levels of C3、C4reduced markedly(P<0.01) in both groups. Conclusion Prophylactic administration of histamine blocker may effectively reduce the side effect of protamine on haemodynamic parameters resulting from the release of histamine, which is caused by protamineactivating the complements in the blood.
