Objective:To detect the expression of Annexin a7and Galectin-3in cervical carcinoma, cervical intraepithelial neoplasia and normal cervix;To clarify the role of Annexin a7and Galectin-3on lymphatic metastasis and clinical prognosis in cervical carcinoma.Methods:Conventional paraffin sections of20normal cervical tissues,45cervical intraepithelial neoplasias and146cervical carcinomas, collected from Pathology Department of Lanzhou University First Hospital from September2009to March2012, were detected by immunohistochemical method (IHC) to assess the expression of Annexin a7and Galectin-3. The statistical analysis of relationship between Annexin a7and Galectin-3expression and all kinds of pathological parameters in cervical tissues were analyzed by χ2test. The correlation between Annexin a7and Galectin-3expression was analyzed by Spearman rank correlation analysis. SPSS20.0package were used for all data analysis.Results:1Positive expression rate of Annexin a7in cervical intraepithelial neoplasia and cervical carcinoma was42.2%(19/45) and69.8%(102/146), respectively. There were significant differences between the two groups (P<0.01), negative expression was observed in normal cervix.2The positive expression rate of Annexin a7protein in cervical adenocarcinoma carcinoma was84.4%(23/26), higher than that in cervical squamous cell carcinoma65%(78/120)(P=0.023); The positive expression rate of Annexin a7in cervical carcinoma with tumor diameter over4cm was88.3%(53/60), significantly higher than that in tumor diameter below4cm57%(49/86)(P=0.0001); The positive expression rate of Annexin a7in deep infiltration was87.5%(56/64), higher than that in the non-deep infiltration (P=3.001); The positive expression rate of Annexin a7in lymph nodes metastasis group was91%(61/67), significantly higher than that in no metastasis group51.9%(41/79)(P=0.0001). Expression of Annexin a7was not related to the age and differentiation of cervical carcinoma(.P>0.05).3The expression rate of Galectin-3protein in cervical carcinoma66%(97/146) was higher than that in cervical intraepithelial neoplasia48.8%(22/45) and normal cervix15%(3/20), the difference was statistically significant in pairwise comparison(P<0.01). The expression rate of Galectin-3protein in cervical adenocarcinoma84.6%(22/26) was higher than that in cervical squamous cell carcinoma62.5%(75/120), the difference was statistically significant (P<0.05).4Galectin3protein was expressed in12cases of poorly differentiated (100%),52cases of moderately differentiated (89.7%),33cases of well-differentiated (66%)cervical squamous cell carcinoma, the difference was statistically significant in pairwise comparison (P<0.01). Positive expression rate of Galectin-3in cervical intraepithelial neoplasia Ⅰ,Ⅱ and Ⅲ was13.3%,46.6%and66.6%, the difference was statistically significant in pairwise comparison respectively (P <0.05).5The positive expression rate of Galectin-3protein with lymph nodes metastasis was90.4%(47/67), significantly higher than that in non-metastasis group53.2%(50/79),the difference is statistically significant (P=0.019); The positive expression rate of Annexin a7in deep myometrial invasion group of was85.9%(55/64), higher than that no deep myometrial invasion of positive expression rate51.2%(42/82), the difference is statistically significant (P=0.001); Galectin-3expression were not related to age and tumor size (P>0.05).6The expression of Annexin a7protein and Galectin-3in cervical carcinoma was positively correlated (r=0.545, P=0.000).Conclusion:1The expression of Annexin a7protein in normal cervical tissue, cervical intraepithelial neoplasia and cervical carcinoma was increased in sequence, suggesting that Annexin a7protein may be associated with the occurrence of cervical carcinoma.2Annexin A7protein expression was related to cervical invasive depth and lymph node metastasis, speculating that the excessive expression of Annexin a7protein is closely related with the progress and prognosis of cervical cancer. Monitoring Annexin a7protein can be used to determine the prognosis of cervical cancer.3Galectin-3might be an important protein in the progress of cervical carcinoma metastasis, Galectin-3might be involved in the occurrence of cervical adenocarcinoma via various mechanisms and other factors, might also be associated with local invasion, metastasis and prognosis of cervical carcinoma.4The expression of Annexin a7protein and Galectin-3protein in cervical cancer was positively correlated, suggesting that both of them might have synergistic effect in the progress of invasion and metastasis of cervical carcinoma.
