Objectives:Candida albicans, Candida tropicalis, Candida krusei and Candida glabrata standard strains were cultured respectively and detect the extracelluar metabolite of the four Candidas.Through detecting the different periods of the four Candida metabolites and drowing the characteristic metabolic profile of each therm as the four candidas standard spectra,and provide a basis for the NMR detecting of Candida in future.To explor the possibility of metabonomics detection method based on1H-NMR in rapid identification of oral Candidas and analysis of the chemical substances of Candida-related metabolites.Analysis of different Candida metabolites and classification of chemical substances, and to explore the influence of different metabolites oral candidiasis.Methods:1.Candida albicans, Candida tropicalis, Candida krusei and Candida glabrata were cultured respectively onto Sabouraud medium.The nephelometry count and growth curve was determined periodically when OD600absorbance values.2.Select the four Candidas broth of the fastest growth period as the NMR detecting sample and draw the four Candidas respective NMR spectra.3.Based on PCA analysis method of four Candida metabolites, and then grouped together in the process, analyze the respective aggregation in the different Candida metabolites in the principal component score plot.4.Relevant metabolic components were finally drawn by comparing with Webspectra database.Results:The four Candida extracellular metabolites had a respective class of clustering relationship while the PCA analysis could differentiate among the four kinds of Candidas.The four Candida metabolomics had different chemical substances which may be associated with different Candida strains.Conclusion: The metabonomics detection method based on1H-NMR is a potential classable method to quickly identify the oral Candidas.The related metabolites analysis also had a good application prospect.
