Objective:To detect the expression of the receptor of inhibin in placental tissue of fetal growthrestriction(FGR) patients and controls to investigate their relationship to FGR, and todiscuss what are the roles of the receptor of inhibin in FGR.Methods:Thirty cases of pregnant women with FGR were selected and twenty cases ofpregnant women which had normal birth weight were randomly selected as control in ourhospital from October2010to December2011.1. HE staining: To prepare paraffin sections of placental samples, observed them underhigh power lens of light microscope after HE staining, to evaluate thesyncytiotrophoblast and interstitial fibrosis, vascular, calcification and necrosis ofvillus.2. Immunohistochemical staining: To prepare paraffin sections of placental samples,using specific antibody of the receptor of inhibin to proceed immunohistochemicalstain respectively, observed their expression under high power lens of light microscopeand half-quantified their expression.3. Realtime PCR: Extracted RNA from homogenate of placental samples, and thenreversed transcription to be cDNA.Finally we acquired two groups of data of thereceptor of inhibin through realtime PCR, and then quantified the relative expression ofthem. Results:1. HE staining slides results: In control group, the villus developping well, whichcontained more blood vessels and syncytiotrophoblast predominated than FGR group,besides, there was more interstitial fibrosis, necrosis and calcification of villus, thecapillary wall was thicker, lumen was more constrictive even occlusive in the controlgroup than FGR group.2. Immunohistochemical staining slides results: In both groups, β-glycan and inhibinbinding protein both express mostly in the cytoplasm of syncytiotrophoblast andtrophoblast, and blood vessel endothelium came second. The degree of positive stain ofβ-glycan in FGR group was significantly higher than control group;but inhibin bindingprotein expression with almost same levels in two groups.3. Realtime PCR results: The quantified relative expression ofβ-glycan gene in FGR groupwas significantly higer than control group, but the quantified relative expression ofinhibin binding protein gene was almost same levels in two groups.Conclusion:1. The expression level of β-glycan in placenta of pregnancy with FGR were obviouslyhiger than normal, while the expression level of inhibin binding protein was almostsame levels in two groups.2. β-glycan cooperate with inhibin was involved in the regulation of fetal growth, thedisorder of this system could result in FGR.3. The expression of β-glycan was greatly improved in late pregnancy, may be play animportant role in the pathogenic causes of FGR.
