Objective: To observe the effects of intermedin on proliferation and apoptosis ofMCF-7breast cancer cells.Methods:[1]MCF-7cells cultured in vitro were randomly divided into control group, IMDgroup and blocker group. control group: the culture medium without any irritants;IMD group: IMD (10-6mol/L); blocker group: AM22-52(10-6mol/L)+CGRP8-37(10-6mol/L).[2]After each group were incubated for24hours, immunohistochemical method wasused to detect the expression of IMD protein,The effect of MCF-7on cellproliferation was detected by methyl thiazol tetrazolium(MTT) assay. Flowcytometry(FCM) was used to observe the cell apoptosis. and detect the expression ofVEGF、COX-2、Bcl-2.Results:(1)The expression of IMD protein in breast cancer MCF-7cells:immunohistochemistry results show the IMD positive expression in MCF-7cells.(2) The effect of IMD on MCF-7cells proliferation: Compared with the controlgroup, MTT experimental results show the IMD can promote cell proliferation,while adding receptor blockers can inhibit cell proliferation (P <0.05).(3) The effect of IMD on MCF-7cells apoptosis: Compared with the control group,the results of flow cytometry show decrease of the cells apoptosis on IMD group, but in receptor blocker group cell apoptosis increased (P <0.05).(4) The expression of VEGF、COX-2、Bcl-2on IMD cells: compared with the controlgroup,the expression of VEGF, COX-2, Bcl-2increase (p <0.05). but in receptorblocker group the expression of VEGF, COX-2and Bcl-2decrease (P <0.05).Conclusion:When treated with IMD in breast cancer cells cultured in vitro it can promote theexpression of angiogenesis-related factor VEGF, COX-2and anti-apoptotic proteinBcl-2increased, and when treated with receptor blocker it can inhibit the expressionof VEGF,COX-2and Bcl-2. It is show that IMD can promote proliferation andmetastasis of human breast cancer through affect the expression of VEGF, COX-2,Bcl-2, suggesting that blocking or down-regulated expression of the IMD toinhibiting or regulation the formation intratumoral microvessel and to promoteapoptosis of the tumor cells, may play an anti-tumor effect, provide a new strategyfor the treatment of breast cancer.
