Objective and significance colon cancer is a commom gastrointestinal tumor in our country, the incidence of the disease has increased year by year.lymphatic metastasis is one of the main way to the transfer, the invasion and metastasis of cancer cell is the leading cause of death in patients.For a long time,chemical treatment, as an important part of colon cancer comprehensive treatment, the treatment effect has been questioned. According to the related research,thymidylate synthase (TS) is one of the key enzymes in fluorouracil drug metabolism.We use immunohistochemical method to detect the expression level of thymidylate synthase (TS) in colon cancer tissue, lymph nodes and normal colon mucosa tissues in order to investigate the correlations of TS to the sensitivity of5-FU chemotherapy and clinicalpathological factors.Methods The samples are collected from General Surgery Department in Second Hospital of Shan Dong University,and after surgical removal of the tumor,it proves a colonic adenocarcinoma.In40cases of patients with colonic adenocarcinoma,24cases of male,16cases of women,aged31—72years old. Among the40cases,the well-differentiated adenocarcinoma are13cases,15patients with middle-differentiated adenocarcinoma,and12patients with poorly-differentiated adenocarcinoma.1case Dukes A period,5cases Dukes B period,27cases Dukes C period,7cases Dukes D period.And among the16cases lymph nodes, there are11cases metastasis lymph nodes and without metastasis lymph node metastasis are5cases. The other40normal colonic mucosa is the control group. We use immunohistochemical method to detect the expression level of thymidylate synthase (TS) in colon cancer tissue, lymph nodes and normal colon mucosa tissues in order to investigate the correlations of TS to the sensitivity of5-FU chemotherapy and clinicalpathological factors.Result1.TS expression in colon cancer tissue are significantly higher than that of normal colon mucosa tissue,Positive rate respectively is,67.5%(27/40)15%(6/40)2.TS positive expression is detected in27of40cases(5cases well-differentiated,10cases middle-differentiated and12cases poorly- differentiated),Positive rate is,38.5%(5/13)>66.7%(10/15、100%(12/12),the total positive rate is67.5%.TS expression in well-differentiated adenocarcinoma is significantly lower than that of the low differentiated adenocarcinoma.3.There are no TS expression in6cases Dukes A,B period.There are20TS expressions in27cases Dukes C period.There are7TS expressions in7cases Dukes D period.Positive rate respectively is,74.1%(20/27)、100%(7/7), the total positive rate is79.4%(27/34).TS expression levels are significantly associated with colon cancer Duke period.4.TS positive expression in12of16cases lymph nodes(there are10of11cases metastasis lymph nodes,2of5cases without metastasis lymph nodes), the positive rate respectively is90.9%(10/11)、40%(2/5),the total positive rate is75%(12/16). TS expression level in colon cancer tissue is positively correlated with lymph node metastasis.5.there is no significant correlation between TS expression and gender, age.Conclusion colon cancer is a commom gastrointestinal tumor in our country, the incidence of the disease has increased year by year.According to the related research,TS expression in colon cancer tissue are significantly higher than that of normal colon mucosa tissue.TS expression is closely related with the incidence of colon cancer,and it is also associated with differentiation of the adenocarcinoma, tumor period and lymph node metastasis. So TS expression level in colon cancer tissue in patients has certain guiding significance to the evaluation and treatment of the colon cancer.
