Study Of Repairition Of Degenerationrabbit Intervertebral Disc ADSCs-PRP Complex

Object: To observe the repair effect of injection of autologous platelet rich plasma (PRP)-adipose mesenchymal stem cells (ADSCs) complexfor degeneration intervertebral disc model.Methods: The preparation of intervertebral disc degeneration model are made by disc puncture，use New Zealand white rabbit.The rabbits aredivided into control group, PRP group, ADSCs-PRPcomplex group randomly.The arterial blood was extracted from New Zealand rabbits to prepare theplatelet-rich plasma by means of two centrifugation. Take interscapular area fat of rabbit to separateand culture ADSCs，then prepare ADSCs-PRP complex. Two weeks later after accomplish ofdegeneration model preparation，injecte PRP and PRP-ADSCs complex to degeneration intervertebraldisc model,according to the group.Control group without injection.Eight weeks later，observe thechange of intervertebral space by X ray，observe the degree of disc degeneration by MRI.Evaluate thesituation of extracellular matrix, and the proliferation and differentiation of nucleus pulposus cells andadipose mesenchymal stem cell by HE stain.Observate the ultrastructure of nucleus pulposus byscanning electron microscope.Results:The bradner disc index (BDI) in the control group, PRP group, ADSCs-PRP groupwere：0.0318±0.005，0.0455±0.007and0.0590±0.008（P>0.05）；the disc height index (DHI) inthe control group, PRP group, ADSCs-PRP group were：46.7%,66.8%and86.6%（P>0.05）. In thecomparison of MRI, increased signal t2-weighted images within the nucleus pulposus can be seen inADSCs-PRP group and PRP group than control group, the former is more apparent..The form ADSCsand the original nucleus pulposus cells are difference observed by Scanning electron microscopy.Conclusion: Injection of PRP and PRP-ADSCs complex can reduce the influence of the discdegeneration, the effect of ADSCs-PRP complex is more outstanding.
