Objective To study the influence of different concentration astragalus membranaceus injection alone or with cisplatin on HeLacell growth,and the preliminary discussion on the action mechanism,and make preliminary discussion for the effect mechanism.Methods Respectively or combined application of different concentrations of astragalus membranaceus injection,cisplatin act on hμman cervicalcancer HeLa cell lines,using3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazoliμm bromide(MTT) method to detect cell growth inhibition,flow cytometric to detect apoptosisrate,Western blot to test expression of apoptosis related protein p53,Bax,Bcl-2.Results1. MTT method detect to cell growth:(1) Different concentration astragalus membranaceus group compared with controls:24h,optical density value of high concentration astragalus membranaceus group was lower than the control group, low concentration astragalus membranaceus group was higher than the control group, the difference was statisticallysignificant (P <0.05), difference between middling concentration astragalus membranaceus group and control group was not statistically significant (P>0.05).48h and72h,optical densi ty value of high concentration astragalus membranaceus group and middling concentration astragalus membranaceus group were lower than control group, low concentration astragalus membranaceus group was higher than the control group, the differences werestatistically significant (P<0.05).(2)Comparison between different concentration astragalus membranaceus groups:24h,48h and72h,optical density value of high concentration astragalus membranaceus groups were lower than middling concentration astragalus membranaceus groups and low concentration astragalus membranaceus groups,middling concentration astragalus membranaceusgroups were lower than low concentration astragalus membranaceus groups,differences were statistically significant (P<0.05).Thus the best concentration of astragalus membranaceus was1000g·L-1.(3)Comparison between different time in same concentration astragalus membranaceus group:(1) The control group: optical density value at48h and72h were higher than24h, the differences were statistically significant (P<0.05);difference between72h and48h was not statistically significant (P>0.05).(2)The high concentration astragalus membranaceus group: opticaldensity value at48h and72h were lower than24h, the differences were statistically significant (P<0.05); difference between72h and48h was not statistically significant (P>0.05). Thus the best time was48h.(3) The middling concentration astragalus membranaceus group: optical density value at72h was lower than24h and48h, the differences were statistically significant (P<0.05);difference between24h and48h was not statistically significant (P>0.05).(4) The low concentration astragalus membranaceus group: optical density value at48h and72h were higher th an24h,72h were higher than48h,the differences were statistically significant (P<0.05).2.Flow cytometry instrμment detecting cell apoptosis rate: control group, astragalus injection group, cisplatin group, astragalus membranaceus injection combined cisplatin group HeLa cell apoptosis rate (%)were为1.00±0.14、2.69±0.21、12.5±0.18and16.8±0.26,the difference between the groups were statistically significant (P<0.05).3.Western blot detecting apoptosis related protein p53, Bax, Bcl-2: Bax, p53protein expression quantity: astragalus membranaceus injection combined cisplatin group was the highest, cisplatin group was the second, blank group was minimμm; Bcl-2protein expression quantity: blank group was the highest, astragalus injection group was the second, astragalus membranaceus injection combined cisplatin group minimμm. The same protein expression between groupgroups all have significant difference (P<0.05). Conclusion higher concentration of astragalus membranaceus injection on HeLa cells have apoptosis-promoting effect,can coordinate cisplatin enhance its apoptosis-promoting effect,which action mechanism is related to enhancing p53,Bax protein epressions andreducing Bcl-2protein expression.
