Objective:Through the establishment of isoproterenol (ISO) ischemicchronic myocardial ischemia rat model to investigate the effects ofTrichosanthes Kirilowii Maxim(TK)and Allium macrostemon Bge (AM)on chronic myocardial ischemia. Explore Trichosanthes Kirilowii Maximand Allium macrostemon Bge on myocardial protective effect andmechanism of ischemic injury.Methods: Manufacture the models of chronic myocardial ischemia byintraperitoneal injection of isoproterenol, and recorded the EGG ofbefore modeling, after modeling, observe changes of electrocardiogramST segment.After two weeks, breakage of rat blood sampling,measuring levels of lactate dehydrogenase (LDH), creatine kinase(CK)and creatine kinase-MB(CK-MB).HE staining of the heart tissue ofrats.Observe myocardial histopathological changes by electronmicroscopy. Western blotting method to prove effects of TrichosanthesKirilowii Maxim and Allium macrostemon Bge on mitogen-activatedprotein kinase (MAPKs) family of3major signaling molecule c-Junn-Terminal kinase (JNK),extracellular signal-regulated kinase (ERK),phosphorylation of P38.Result: The TKAM can effectively reduce ST segment elevation,(P<0.01);The model group compared with normal group, increase serum CK-MB、LDH、CK activity, TKAM can lower levels of CK-MB、LDH、CK.TKAM can improve the myocardial fibers disorder,vacuolar,organelle swelling which caused by ischemia. P-P38,P-JNK,P-ERK,MAPK protein levels of Ischemia myocardial tissueincreased significantly, TKAM can effectively lower levels ofP-P38,P-JNK,P-ERK.(P<0.01)Conclusion: TKAM has protective effect on chronic myocardialischemia. And TKAM are better than TK or AM alone. The mechanismmay be adopted on scavenging oxygen free radicals, reduce theoxidation inhibition of P38、JNK and ERK phosphorylation of proteins inorder to reduce ischemic injury.
