Ampelopsis megalophylla Diels et.Gilg is a medicinal plant of Ampelopsis.It is a herb commonly used in Enshi of Hubei province. It is neutral innature,tasting sour and astringent and has an effect of removing pathogenicheat from blood. Pharmacological studies suggested that the Ampelopsismegalophylla extracts had the effect of antihypertension and antivirus.This article summarized the study status of the fingerprints and the serumpharmacochemistry research.At the same time, this article made a review andsummary of the damage to target organ caused by hypertension and theprotective effect on target organ because of antihypertensive drug.It laid afoundation for our experiments.The optimal preparation technology, which was determined in previousresearch, was adopted to preparating flavonoid extract from Ampelopsismegalophylla. HPLC was applied to measure and analyse fingerprints ofeleven extract samples of medicinal materials from West Hubei、Hunan andChongqing Province. Specific chromatographic conditions were as follows:the components were separated using an Agilent TC-C18column(4.6mm×250mm,5μm), gradiently eluted with the mixture of acetonitrileand0.2%phosphoric acid aqueous at30℃, and monitored with UV detector at265nm, and sample volume injected was20μL. The HPLC fingerprints showten common peaks and four characteristic peaks were identified(Ampelopsin、 Myricetin、 Myricitrin and Quercetin).The result was in accordance with thetechnical requirement of the chromatography fingerprints.This HPLCfingerprints provided a basis for the controlling the quality and the reserch toserum pharmacochemistry of the flavonoid extrat of Ampelopsismegalophylla.Take rats as research object,prepare medicated serum by the way of serumpharmacology of traditional Chinese medicine and establish the chromatogramof serum pharmaeoehemical ingredients by HPLC. By comparing thechromatogram of drug serum to the chromatogram of flavonoid extrat fromAmpelopsis megalophylla, the result indicated that there were threecomponents absorbed into blood, one was Ampelopsin,the other two neededfurther study. The result also indicated that the composition absorbed intoblood in a high proportion was the composition which had a high level inflavonoid extract from Ampelopsis mealophylla. This method was used for thefirst time and it was stable、dependable and reproducible.The chromatogramcan show all the changes of ingredients of the flavonoid extract fromAmpelopsis mealophylla before and after absorption into blood. It laid afoundation for further reserch to certainment of pharmacodynamic substanceand metabolism mechanism.SHR models were chosen to study protective effect flavonoid extract fromAmpelopsis mealophylla on heart and kidney.Experimental study showed thatmedium and low groups of flavonoid can observably decrease the level ofUcr、Urine Protein and Scr.Medium groups can significantly increase Vol of24h and decrease BUN. The level of Ccr of medium and low groups has atendency to increase. Pathological research showed that medium and lowgroups of flavonoid extract can reduce the damage and renal fibrosis ofSHR,but the effect of it on heart was not sure. Through this reserch, it wasdetermined that medium and low groups of flavonoid extract probably had good protective effect on renal and more researches needed to be done tostudy the effect on heart....
