| Background: Diabetic nephropathy (DN) is one of the common clinical refractory chronic complications of diabetes, glomerulosclerosis late outcome of a variety of causes glomerular injury or disease,and how to prevent and delay glomerulosclerosis people urgently solve the problem. Nephritis, the rehabilitation piece has anti-inflammatory, renal glomerulosclerrosis progress and reduce the role of diabetic kidney damage, but the exact mechanism is unclear.Objective: Observed the impact of the Shenyankangfu tablet to glomerulosclerosis in diabetic rats and affect on BMP-7/TGF-β1/Smadl signal transduction pathway. Investigate the mechanisms of the Shenyankangfu tablet on the protective of diabetic kidney damage.Methods: To male34SD rats were randomly divided into normal contact (NC group), diabetic contact (DC group) and the Shenyankangfu tablet magnets group (SYK group). Application of streptozotocin(STZ)60mg/kg intraperitoneal injection,to establish of animal models of typel diabetic nephropathy.The SYK group rats given Shenyankangfu tablet according to600mg·kg-1·d-1dose (soluble in1%sodium carboxymethyl cellulose), NC group and DC group given an equivalent dose to normal saline,rats were sacrificed after8weeks. Determine blood glucose (BG), Glycosylated hemoglobin (HbAIC), serum creatinine (Scr), blood urea nitrogen (BUN),24hour urinary protein (24hU-TP), calculate the kidney weight index (mg·g-1); specimens from kidney samples detected by immunohistochemical method the kidney tissues. Observe the expression of BMP-7、TGF-pβ1、Smad1、Col-Ⅳ and CTGF.Results:1. Compared with NC group, DC group and SYK group rats showed apathetic,unresponsive dull hair,and polydipsia,urine output increased significantly (P<0.01), weight loss (P<0.01) and the kidney weight index (P<0.01). SYK group rats varying degrees of improvement compared with DC group (P<0.01).2. Compared with NC group, the SYK group and DC group was significantly increased in24hour urinary protein, serum creatinineand blood urea nitrogen, a random blood glucose and blood urea nitrogen glycosylated hemoglobin levels was significantly higher (P<0.01or P<0.05). Compared with DC group, the SYK group24hour urinary protein, serum creatinineand blood urea nitrogen, decreased (P<0.05); random blood glucose and blood urea nitrogen glycosylated hemoglobin levels was not statistically significant (P>0.05).3. HE stained, observation by light microscopy:Glomerular structural integrity of NC group, the glomerular capillary rich, thin wall, the lumen open within red blood cells. Glomerular mesangial area showed no abnormal change, did not alter the mean glomerular area and volume. The DC group glomerular structure damage, wall thickening luminal narrowing. Glomerular mesangial area widened, mean glomerular area and volume increased. The difference was statistically significant (P<0.01). Compared with DC group, the SYK group reduced the degree of thickening of the glomerular mesangial area, capillary wall thinning, the capillary cavity open, mean area and volume of glomerular decreased, the difference was statistically significant (P <0.01).4. Immunohistochemical observation of BMP-7、TGF-β1、Smadl、Col-Ⅳ、CTGF protein expression:Compared with NC group, the DC group rats glomerular BMP-7protein expression decreased significantly (P<0.01), and TGF-β1, Smadl, CTGF and Col-Ⅳ protein expression in glomerular increased significantly (P<0.01). Compared with DC group, the SYK group rats glomerular BMP-7protein expression was increased (P<0.01), TGF-βi, Smadl, CTGF and Col-IV protein expression decreased significantly, the difference was statistically significant (P<0.05).Conclusion:1. The Shenyankangfu tablet can reduce diabetic rats, capable blood serum creatinine and blood urea nitrogen levels, reduce urine protein, to reduce glomerular hypertrophy, and improve kidney function.2. The Shenyankangfu tablet can inhibit accumulation of CTGF and Col-Ⅳ in diabetic rat, increase in BMP-7protein expression and reduce the expression of TGF-β1/Smadl, delay glomerulosclerosis reduce kidney damage. |
