Objective Gastric cancer is one of the most common cancers in humans,gastrointestinal hormone plays an important role in its complexdevelopment process. As a kind of brain-gut peptide, Bombesin(BN)widely distribut in the P cells which plays an important role on thedigestive function mostly in gastric antrum and duodenum. BN is abioactive peptide containing14amino acid residues and its receptor isG-protein coupled receptors. To autocrine or paracrine growth factorpromotes the proliferation of various types of cells, especially tumor cells.But it is not clear that dynamic changes and mechanism of BN in theevolution of gastric precancerous diseases (lesion) and gastric cancer.In this study, we used immunohistochemical SP and RT-PCR todetermine the expression of BN in chronic superficial gastritis, chronicatrophic gastritis, atypical hyperplasia and gastric cancer. Investigate thevalue of BN as a biological marker in gastric cancer and gastricprecancerous diseases. Establish the experimental basis for monitoringand diagnosis of gastric precancerous diseases (lesion) and gastric cancerin the clinical work. Lay the theoretical foundation of a new way thattreats gastric cancer with the antagonist of BN.Methods This study collected the biopsy specimens of patients receivedgastroscopy and the resected specimens of parts of the surgery patients inFengtian Hospital of Shenyang Medical College from May2011to May2012, including28cases of chronic superficial gastritis,21cases ofchronic atrophic gastritis,17cases of gastric mucosal atypical hyperplasiaand18cases of gastric cancer. All cases were confirmed by HE staining, and gastric cancer caseswere not treated with radiochemotherapy. Immunohistochemical SP andRT-PCR determined the expression and distribution of the protein andmRNA of BN respectively in the groups of gastric mucosa in order tofurther explore the mechanism of the occurrence and development ofgastric cancer. SPSS13.0statistical analysis software to analysis theexperimental data, all data signed in mean±standard deviation (X±SD),single factor analysis of variance (ANOVA) and Bonferroni test wereused for multiple group, P <0.05has significant difference.Results Hematoxylin and eosin (HE) staining pathological diagnosis ofgastric mucosa:28cases of chronic superficial gastritis,21cases ofchronic atrophic gastritis,17cases of gastric mucosal atypical hyperplasia,18cases of gastric cancer. Immunohistochemistry SP to determine theexpression of BN: distribution of BN immunoreactive particles is brown,the staining in the cell membrane and cytoplasm, nucleus withoutcoloring. Chronic superficial gastritis, chronic atrophic gastritis, gastricmucosal atypical hyperplasia and gastric cancer group, the averageoptical density values were (0.1318±0.0371),(0.1114±0.0194),(0.3387±0.0465),(0.3946±0.0621). Compared with two groups, group ofgastric cancer and atypical hyperplasia group were significantly higherthan that of chronic superficial gastritis and chronic atrophic gastritisgroup, there were statistically significant differences. But betweenatypical hyperplasia group and gastric cancer group, there was nosignificant difference between groups, and that of chronic superficialgastritis group and chronic atrophic gastritis group. RT-PCR to determinemRNA expression of BN: expression of mRNA in chronic superficialgastritis, chronic atrophic gastritis, gastric mucosal atypical hyperplasiaand gastric cancer decreased firstly and then increased, The ratio of IDV between the ratio of IDV of BN to IDV of β-actin (0.1100±0.0371),(0.0914±0.0213),(0.3017±0.0625),(0.6958±0.0724), respectively.There are significant differences in all groups which compared with eachother except for chronic superficial gastritis and chronic atrophic gastritisgroup.Conclusions In the process of gastric carcinogenesis, the expression ofprotein and mRNA of BN showed a gradually increased tendency, and itprompted BN may have a certain role in the occurrence and developmentof gastric cancer and precancerous lesions.BN plays a catalytic role in theevolution process with the effect of prompting and monitoring in differentstages of disease.
