Marine natural products with diverse chemical structures and various biologicalactivities attracted a great attention for the marine-derived new drug development.This thesis mainly focus on the isolation and the structural identification of soft coralScleronephthya sp.Using extensive chromatographic separation including silica gel, ODS, SephadexLH-20and semi-preparative HPLC, ten compounds were isolated. Their structureswere identical to pregna-1,20-dien-3-one, pregna-1-methoxy-20-en-3-one, pregna-1,4,20-trien-3-one, pregna-1-en-3-one-20β-O-acetate, pregna-1-en-3-one-20α-O-acetate, pregna-20-en-3β-ol, Pregna-5,20-dien-3β-ol, pregna-1,7-dien-20-ol-3-one,pregna-1-en-20-ol-3-one and pregna-1,4-dien-20α-ol-3-one. Using C. elegans Daf-4(m63) mutant model, the MeOH extract and pregna-20-en-3β-ol showed inhibitoryactivity. This result provides research base for further rational use and thedevelopment of soft coral Scleronephthya sp. |
