Lymphoma, a common malignant hemotopoietic tumor, isoriginated from the lymphoreticular system, the incidence of the diseaseis on the rise in our country recently. The conventional therapeuticapproaches for lymphoma are operation, radiotherapy, chemotherapy,etc.But most of them have limited therapeutic effects and seriousside-effects. Recently, more and more biological therapeutic strategieshave been used for lymphoma, such as the rational use of monoclonalantibodies or combined with conventional chemotherapy drugs canimprove the therapeutic effects, reduce adverse reactions and improvethe long-term survival rate of lymphoma patients. Rituximab is one kindof monoclonal antibodies, it has significant therapeutic effect in clinicaltreatment of NHL, but there are still some patients have primary orsecondary resistance to rituximab, so we need to find a more effectivemethod to prolong the survival period of time.To study the role of rituximab in inhibiting Raji cells and NK cells,the proliferation of Raji cells and NK cells were detected by MTTmethod; cytotoxicity aganist Raji cells of NK cells was detected by CCK8method respectively before and after treated by rituximab, and thedifference of killing effects of NK cells, γδT cells and combination cellson Jurkat cells were detected by CCK8method respectively.The study showed that Raji cells were inhibited by rituximab, theinhibition effecte was increased with recreation time; we successfullycultured and amplified high purity of NK cells and γδT cells from theperipheral blood of lymphoma patients, and the killing effect of NK cellson Raji cells was significantly increased with effector to target ratio invitro; the killing effect of NK cells on Raji cells was increasedsignificantly after combined with rituximab. and the killing effect of NKcells combined with γδT cells on Jurkat cells was more significant thanNK cells or γδT cells, both outcomes showed a synergistic effect.The conclusions of this study are as follow:1. Rituximab inhibits Raji cells, the growth inhibitory rate ofrituximab on Raji cells is associated with recreation time;2. We successfully culture high purity of NK cells and γδT cells, thekilling effect of NK cells on Raji cells is significant and in adose-dependent manner;3. There is a synergistic killing effect of NK cells combined withrituximab on Raji cells;4. There is a synergistic killing effect of NK cells combined withγδT cells on Jurkat cells.
