ObjectiveTo compare cardioprotective effects of trimetazidine and ischemic post-conditionings in rats on ischemia-Reperfusion Injury.MethodsThe60Wistar rats, half male half female, were divided into4groups randomlyby weight, with15rats for each group:⑴Sham-operated group: It was only treated by opening chest and heart withoutligation.⑵Model group: It was treated for LAD occlusion after chest opening, resultingin ischemia of30min; then release thread tension to recover the coronaryflow for120min.⑶Trimetazidine group: Intragastric administration of10mg/kg trimetazidinewas given before24h and60min of reperfusion, ascertain peak of bloodconcentration.⑷Ischemic post-conditioning group: It was unfastened for30s reperfusion and30s religation after ischemia for30min, and a120min reperfusion was givenagain after three times circulation.The rat abdominal aorta blood was taken for each group, and after standing andcentrifugation, the supernatant was taken to measure the activities of serumsuperoxide dismutases (SODs), malondialdehyde (MDA), creatine kinase (CK), lacticdehydrogenase (LDH) and aspartate transaminase (AST);The rats of each group were removed for heart, of which,5of them were used tomeasure the myocardial infarction size,5were used for myocardial microstructureobservation with light microscope and electron microscope; the last5were examinedfor expressions of bcl-2protein and caspase.ResultsCompared with the Model group, Myocardial infarction size of theTrimetazidine group and the ischemic post-conditioning group was reduced (p<0.05);Myocardial injury of both the two conditioning group was alleviated and inflammatory cell infiltration was reduced when observing with light microscope;Cardiac muscle fiber injury and mitochondrial swelling of both the two conditioninggroup were alleviated when observing with electron microscope; CK, LDH and ASTcontents of myocardial enzyme series of both the two conditioning group weredecreased greatly (p<0.05); SOD content in serum of both the two conditioninggroup increased and MDA content reduced (p<0.05, p<0.05); The bcl-2proteincontent in myocardium of both the two conditioning group increased, and caspasecontent decreased (p<0.05, p<0.05). The difference between the Trimetazidinegroup and the post-condition group are not sharp(P>0.05).ConclusionTMZ post-conditioning and ischemic post-conditioning⑴Effectively decreasemyocardial cells death, and reduce the myocardial infarction size;⑵decrease theMDA content in serum and increase the SOD content;⑶May reduce the injury ofmitochondria in myocardial cells, stabilize its structure, while reduce the inflammatory cell infiltration;⑷Alleviate the apoptosis of myocardial cells;Both of the two conditioning methods showed significant protective effects onmyocardial ischemia-reperfusion injury, and there’s no significant difference betweenthe two methods.
