The main reason of adult tooth loss is the alveolar bone resorption.And it is also the mostcommon human bone destructive diseases.Alveolar bone absorbed not only affectsmastication、pronunciation、eating、and other functions,but also affects the patient’s physicaland mental health and quality of life. Mechanism study of alveolar bone absorption and itspathogenesis, finding the reason of inhibiting bone absorption, promoting bone formation,and regulating the microenvironment which have important practical significance onprevention and treatment of alveolar bone resorption.Oligonucleotide MT01is a kind of single DNA nucleotides designed based on DNAsequence of mitochondrial. We confirmed in our previous work, MT01can promoteosteoblast activation、inhibit alveolar bone resorption of rat periodontitis model. Research hasshown that MT01can block the activation of intracellular TLR-9, relieve the inflammatorytissue damage.Based on these researches, we want to research whether MT01can inhibit the activationof TLR-9of osteoblast,and inhibit the expression of cytokines which are assocoated withalveolar bone absorption. We want to detect whether MT01can regulate the expression ofinflammatory cytokines of osteoblast.These ideas could be the experimental basis of MT01may be inhibition of alveolar bone absorption.Objective:The mechanism research of MT01influence the mRNA expression level of TLR-9、IL-1、TNF-αof osteoblast via real-time PCR. To estimate the role of the specific ODNsequences on the regulating bone remodeling.Method:This experiment research the influence of MT01on the mRNA expression level ofTLR-9、IL-1、TNF-αof osteoblast via experiment in vitro. The experiment divided into experimental group and control group.Every group has three wells and was divided into6h、12h、24h、48h、72h. Real-time quantitative PCR were used to analysis the mRNA expressionof TLR-9、IL-1、TNF-αin the experimwntal group and control group. Analysis theexperimwntal data using the One-way ANOVA in the statistical software of SPSS18.0.Results:The results showed that①MT01can significantly reduce the mRNA expression ofTLR-9in the time of6h、48h、72h;②MT01can significantly reduce the mRNA expressionof IL-1in the time of6h、24h、48h、72h;③MT01can significantly reduce the mRNAexpression of TNF-αin the time of24h、72h.Conclusion:The result suggested that MT01could inhibit the mRNA expression level of TLR-9、IL-1、TNF-αof osteoblast. All of these shows that the mechanism of MT01can promote theosteogenesis may be related to the gene expression unusual of inflammatory cytokines.
