The Expression And Significance Of α-SMA In Infarct Foci And Villous Clump Of Basal Plate Of Patients With Severe Preeclampsia

Objectives To detect the expression profiles of alpha-smooth muscle actin (α-SMA)in infarct foci and villous clump area of basal plate and to analyze the relationshipsbetween these and severe preeclampsia (SPE).Methods40women with SPE (n=20for early-onset preeclampsia (EOSPE) andn=20for late-onset preeclampsia (LOSPE)) enrolled in obstetrics of the Jilin UniversityBethune Second Hospital from January2011to March2012were chosen as trial group.38normal pregnancies with matched gestational age (n=18for early control (EC), n=20for late control (LC) were chosen as control. Expression location of α-SMA in basalplate, and relative expression levels of its mRNA and protein were examined byimmunohistochemistry, RT-PCR and Western Blotting, respectively.Results1.The prevalence of multifocal infarct was90.00%in20EOSPE patients,15.00%in20LOSPE patients,5.56%in18EC patients and10.00%in20LC patients,respectively (χ2=48.24, P<0.05). The prevalence of villous clump was EOSPE＞LOSPE＞LC＞EC (75.00%,60.00%,35.00%and11.11%)(χ2 =17.52, P<0.05).αα-SMA was located in smooth muscle cells of villous vessels and myofibroblasts ofvillous stroma of basal plate.3.Expression levels of α-SMA were increased in late, transitional and early multifocalinfarct or multifocal villous clump successively (P<0.05), and were positively correlatedwith the amounts of smooth muscle cells of villous vessels and myofibroblasts of villousstroma in placental bed stained by α-SMA antibody.4. The amounts of stained cells decreased progressively in big, medium and small villiof multifocal infarct or multifocal villous clump.Conclusion The multifocal infarct and multifocal villous clump might participatein the pathogenic progress of SPE through blocking blood vessels and making thevessels contracted and aggravating uterine tension by α-SMA.
