Objective: Gallbladder carcinoma is one common malignant tumorin the biliary diseases, higher malignancy and poor prognosis, thedevelopment of a complex multi-stage process, with a variety of changesin the molecular level related closely. In recent years, a large number ofstudies have shown that overexpression of p-STAT3closely related withdevelopment of gastrointestinal tumor. p-STAT3is a form of STAT3activation status,can quickly shift to the nucleus, and the role of specificDNA binding sites in the nucleus, directly or indirectly, raised with cellproliferation, apoptosis, angiogenesis, invasion and metastasis relatedgenes. The purpose of this experiment is to detect p-STAT3, Bcl-2andMMP-7expression in gallbladder carcinoma and chronic cholecystitisexplore the role in the development of the relationship between threegallbladder carcinoma,and analysis the relationship about them.Methods: Collecting64cases of gallbladder carcinoma patientsafter radical surgical resection pathology specimens from January2000toOctober2012in the First Hospital of Jilin University, as the experimentalgroup. Take over the same period due to chronic cholecystitis,10caseswho gallbladder removed were selected as the control group.Theexpression of p-STAT3and Bcl-2、 MMP-7were examined byimmunohistochemical technique(SP) in64cases of gallbladdercanrcinoma,and10cases of chronic cholecystitis. Analyszed p-STAT3, Bcl-2and MMP-7protein expressions in gallbladder carcinoma andchronic cholecystitis expression differences by statistical analysis, as wellas the relationship with clinical and pathological features.Results: p-STAT3, Bcl-2and of MMP-7positive expression ingallbladder carcinoma were82.8%、79.7%、81.3%, respectively,werehigher than chronic cholecystitis, and the differences were statisticallysignificant (P <0.05). p-STAT3, Bcl-2and MMP-7positive expression ingallbladder carcinoma are related with clinical stage and lymph nodemetastasis (P <0.01), while without the patient’s gender, age and largelyindependent of the degree of differentiation (P>0.05).p-STAT3and Bcl-2protein expression levels were positively related in the tissue ofgallbladder carcinoma (P <0.01). The expression levels of p-STAT3andMMP-7were positively associated among them (P<0.05). The expressionlevels of Bcl-2and MMP-7were even positively associated among them(P <0.01).Conclusion: The Upregulation of p-STAT3, Bcl-2and MMP-7ingallbladder carcinoma may be related to the occurrence and development ofgallbladder cancer. p-STAT3and Bcl-2, MMP-7play synergies in theoccurrence and development of gallbladder carcinoma synergies, to promotegallbladder cancer cell invasion and metastasis; expression of Bcl-2andMMP-7play synergies in the development of gallbladder cancer.
