Objective:To confirm the effects of curcumin on oxidative stress in the co-culture system including Human Fetal Lung Fibroblasts and A549. Discuss the potential and protective mechanism of the prophylactic effect of curcumin on pulmonary fibrosis. And find the specific target of curcumin on oxidative stress.Methods:The Human Fetal Lung Fibroblasts which co-cultured with A549were divided into five groups:control group:the cells were cultured in DMEM without TGF-βl or curcumin. TGF-βlgroup:the cells were cultured in DMEM containing5ng/mL TGF-βl. TGF-P+cucurmin treatment group:the cells were cultured in DMEM containing5ng/mL TGF-βl and different concentration Curcumin(5、 l020μM). ELISA was used to analyse the content of TNF-a in culture supernatant. Serum level of MDA and SOD were tested by Spectrophotometric. Intracellular ROS production was detected by flow cytometry analyse. NF-κB and APE/Ref-1were measured by western blotResults:The serum MDA, Intracellular ROS, the content of TNF-a and the nuclear factor NF-κB protein expression of TGF-βl group were significantly increased while the activity of SOD and APE/Ref-l protein expression were significantly decreased.(p<0.01), it illustrates that the oxidative level of Human Fetal Lung Fibroblasts was obviously increased after TGF-βl stimulated. After curcumin of different concentration intervened, the serum MDA, Intracellular ROS, the content of TNF-a and the nuclear factor NF-κB protein level were significantly decreased while the activity of SOD and the level of APE/Ref-l were obviously increased(p<0.01).Conclusions:Curcumin of low concentration can reduce the oxidative level of Human Fetal Lung Fibroblasts which is cocultured with A549after TGF-βI stimulated, and significantly increase the level of Intracellular anti-oxidative protein APE/Ref-l,it implies that curcumin may intervene pulmonary fibrosis by reduce oxidative level.
