Objective:The immune function disorder of HBV-associated hepatocellular carcinoma patients not only because of tumor cell’s influence but also be HBV infection effect. There are inimitable advantage in which traditional Chinese medicine interfere in immunity adjustment, This study was to establish a immune model of HBV-associated hepatocellular carcinoma in vitro and analyze the effect of traditional Chinese medicine. The proposed approach provides a new idea for the immunoregulation mechanism of HBV-associated hepatocellular carcinoma with the effect of Chinese Traditional Medicine。Method:In this study, extracting PBMCs from the peripheral blood of chronic HBV-associated hepatocellular carcinoma patients with different states,using DCs adherent growth characteristics and magnetic cell separation technology (Magnetic Activated Cell of the Sorting, the MACS), the establishment of hepatoma cell lysates pulsed DCs, T cell interaction model, the use of plasma pharmacology research methods, flow cytometry, DCs surface molecules of CD1a/CD83/HLA -DR/CD80/CD86and T cell surface molecules CD3/CD4/CD8/CD4+CD25+CD1271owTregs receptor expression were observed and compared before and after the intervention of drugs in plasma, peripheral blood immune cell function changes.Results:1.Normal people with HBV-associated hepatocellular carcinoma patients with T-cell phenotype, patients CD3+, CD4+expression rate was significantly lower than normal CD8+expression is high in normal individuals, the difference was statistically significant (P<0.05).Normal subjects and HBV-associated hepatocellular carcinoma patients DC cell phenotype, patients of CD1a, CD83, HLA-DR, CD80and CD86expression was significantly lower than normal, the difference was statistically significant (P<0.05).2. DCs of HBV-associated hepatocellular carcinoma patients pushed with the drug plasma and hepatoma cell lysates compare with DCs with the non-drug plasma and hepatoma cell lysates, CD1a, CD83, HLA-DR, CD80and CD86expression had significantly higher levels,the difference was statistically significant (P<0.05).3. Handled by the drug plasma and hepatoma cell lysates in load DCs incubated T cells compare with that without the drug plasma treatment, the only the hepatoma cell lysates load of DCs incubated T cells,CD3,CD4expression had significantly higher levels and CD8 expression had significantly lower levels. difference was statistically significant (P<0.05). The difference of CD4+CD25+CD1271owTregs expression wasn’t statistically significant (P>0.05).Conclusion:1.HBV-associated hepatocellular carcinoma patients DC there is a mature obstacles, costimulatory low capacity, low T-cell immune function, there is a severe immune dysfunction.2.HBV-associated hepatocellular carcinoma patients through the recovery of DCs function to restore the T lymphocytes function, improved immune function;3. strengthening spleen herbs can restore function of peripheral blood DCs of the immune clearance phase and the immune tolerance HBV-associated hepatocellular carcinoma patients to restore its downstream T lymphocyte function. But it is invalid to regulate the CD4+CD25+CD1271owTregs expression.
