The Experimental Study On The Effects Of Liuweidihuang Decoction On MMP-2and TIMP-2Expression With5/6Nephreetomy Rats

Objective:Observation of Liuweidihuang Decoction on5/6nephrec-tomized rat remnant kidney of,MMP-2and TIMP-2expression influence, discussion of Liuweidihuang Decoction to protect renal function, the mechanism of delaying kidney damage.Methods:In accordance with the completely random methods72male SD rats were divided into5groups:Black group, sham operation group, model group, enalapril group, Liuweidihuang Decoction group. The model group, the enalapril group, Liuweidihuang Decoction group with5/6nephrectomy induced chronic renal failure rat model, Sham operation group undergoing operation two times at the same time,but do not damage kidney.Second operation after3days, the rats were treated by gavage. In eighth weeks in rats were sacrificed, observation of remnant kidney tissue morphological changes were observed with immunohis-tochemistry rat kidney MMP-2and TIMP-2expression.Results:1. By using H-E and Masson staining with5/6nephrectomy rats renal tubular interstitial tissue morphological changes, Liuweidihuang Decoction and enalapril group can reduce renal tubulointerstitial damage and fibrosis degree;2.Immunohistochemical semiquantitative analysis revealed six rehmannia root group of renal cortical MMP-2more than and TIMP-2expression is less than the model group (P＜0.01);3. Liuweidihuang Decoction can lower blood levels of Scr.Conclusion:Liuweidihuang Decoction may be adopted to decrease the accumulation of extracellular matrix, upregulation of MMP-2,downregulation of TIMP-2expression, make the cell matrix components decreased synthesis,in order to reduce inflammation and fibrosis,to prevent renal interstitial fibrosis, protect the kidney function.
