Approximately2billion people present evidence of past or present infection with hepatitis B virus (HBV) over the world. With more than350million chronic HBV carriers, this virus stands as one of the most common human pathogens and it causes significant public health problems. however, has not been fully described. hepatitis B virus X protein(HBx) stimulates and influences signal transduction pathways within cells. The interplay between the viral and host factors activates oncogenic signaling pathways and triggers cell transformation. The various types of genomic changes in HBV-induced HCC-chromosomal instability, telomere attrition, and gene-level mutations. In addition, HBx can promote genomic instability as the main features as viral oncoproteins.The RecQ DNA helicase family is conserved in all organisms and participates in the maintenance of genomic integrity of cells. Microorganisms such as Escherichia coli, Saccharomyces cerevisiae contain only one RecQ helicase, but higher eukaryotes have more than one RecQ helicase. In human cells, the RecQ helicase family comprises RecQLl (also known as RecQL or RecQl), BLM, WRN, RTS (also known as RecQL4), and RecQ5. We showed that these.RecQ helicases are expressed in the cell nucleus, are upregulated with DNA replication, and participate in DNA repair during cell proliferation.To further show the biological function of HBx and its role in hepatocellular carcinoma. in a yeast two-hybrid screening for potential HBx-interacting proteins, we identified a human RecQ protein-like (DNA helicase Q1-like)(RECQL). The interaction was further verified by co-immunoprecipitation experiments in mammalian cell, Immunofluorescent analysis showed that HBx was co-localized with RECQL. To further explore the domain mediating the interaction between HBx and RECQL, several truncated mutants of HBx and RECQL were constructed respectively.our research result is that RECQL can interact with HBx. RECQL can maintain the chromosomal stability, HBx may act the promoting factor by inhibiting DNA repair causing DNA damage and accumulation error, thereby, contributing to the HCC development. In HBx-related HCC, how does the HBx inhibit the chromosomal stability, it is our next work. Our data can promote the chromosomal instability about HBx.
