Study On Antimicrobial Effect Of Chaenomeles Speciosa S. Nakai Crude Extract

Chaenomeles Speciosa S. Nakai was extracted by bain-marie vibration method with95%ethanol and distilled water as solvent in order to study the antimicrobial effect of Chaenomeles Speciosa S. Nakai crude extract and the influence of ultraviolet ray and temperature on antimicrobial effect.After filtering and rejecting filter residue,ethanol filtrate and distilled water were concentrated into extract in rotary evaporator by vacuum concentration and then vacuum drying. Ethanol extract and distilled water extract were prepared as solution whose concentrations were500.00mg/mL by dimethyl sulfoxid and sterile water, respectively. Two different crude extracts were tested the antimicrobial effect to Pseudomonasfluorescens, Escherichia coli, Bacillus subtilis, Staphylocaccus aureus, Micrococcus tetragenus and minimum inhibitory concentration and minimal bactericidal concentration by disk diffused tests. The results showed that95%ethanol crude extract and distilled water extract possessed obvious antimicrobial effect to five harmful bacterias, the sequence of the antimicrobial effect of95%ethanol crude extract is Micrococcus tetragenus Esherichia col、Staphylococcus aureu、Bacillus subfilis、Pseudomonas fluorescens. The sequence of the antimicrobial effect of95%ethanol crude extract is Micrococcus tetragenus、Staphylococcus aureu、Esherichia col、Bacillus subfilis、Pseudomonas fluorescens. Minimum inhibitory concentration of95%ethanol crude extract on five bacterias is0.98mg/mL、3.91mg/mL、15.63mg/mL、1.96mg/mL、7.82mg/mL, respectively.Minimum bactericidal concentration of95%ethanol crude extract on five bacterias is3.91mg/mL、3.91mg/mL、31.25mg/mL、3.91mg/mL、7.82mg/mL, respectively; Minimum inhibitory concentration of distilled water crude extract on five bacterias is62.50mg/mL、15.63mg/mL、31.25mg/mL、15.63mg/mL15.63mg/mL, respectively; Minimum bactericidal concentration of distilled water crude extract on five bacterias is62.50mg/mL、15.63mg/mL、62.50mg/mL15.63mg/mL、15.63mg/mL, repectively. What’s more,95%ethanol crude extract of antimicrobial effect was superior to that of distilled water extract,crude extracts both had good ultraviolet stabilization and thermo stabilization.
