Construction Of DNA Vaccine About Candida Albicans HSP90Epitope C And Evaluation On Its Immunological Activity

Candida albicans and other Candida species are opportunistic yeast, found in thegastrointestinal、oral cavity and reproductive mucosa of healthy individuals. However, inimmunocompromised settings, it can increase and transform into a pathogenic killer, leadingto oral and oropharyngeal,mucocutanneous candidiasis(Mucosal infections), evenlyspreading into the bloodstream and internal organs (Systemic infection).Treatment of C.albicans infection remains reliant on antifungal drugs, but in the use ofantifungal drugs, the disease’s mortality rate remains as high as70%.The best way of controlC.albicans is preventing infection. Hence, vaccine, capable of stimulating immunity, wouldbe considerable value. DNA vaccine,compared with traditional vaccine, contains efficient,inexpensive, safe and many other advantages. Progress in this field has resulted in thedevelopment and marketing of three veterinary DNA vaccine since2005.In unfavorable conditions, C.albicans can secrete heat shock protein90(HSP90), andHSP90is also an immuneodominant antigen of Candida albicans. Matthews and othersidentified the epitope map of C.albicans HSP90.There are three common continuous epitopesin the end of HSP90: epitope C、epitope B and epitope H. But,they proved that only epitopeC and epitope H have immunogenicity.In this paper, we choose the gene sequence that encodes the HSP90’s epitope C ofC.albicans and insert the sequence into the prokaryotic plasmid pET28a and eukaryoticplasmid pcDNA3.1(-). We obtain two kind of recombinant plasmid: pET-28a/HSP90C andpD-HSP90C.The recombinant protein is expressed of pET-28a/HSP90C in Ecoli. Accordingto text of Western Blot,we have proved the recombinant protein’s immunogenicity with theserum of cancer patients and immunized mice by recombinant protein. pD-HSP90C is socalled DNA vaccine. We immune the mice by way of intramuscular injection and electronictransfer with DNA vaccine, getting some immuned serum. Through the test of WesternBlot,we prove the DNA vaccine ’s immunogenicity of C.albicans. These results founde thefurther research DNA vaccine of C.albicans.
