ObjectiveThrough the experimental study of proliferation and differentiation of rabbitbone marrow mesenchymal cells on PRF membrane,to research the role offrom MSCs to osteoblast what PRF do in the Transformation process.theobjective is providing an evidence in the study of PRF on the Oral implantology.MethodsUse the femur and tibia of seven days’rabbit,separate into Bonemesenchymal stem cells to open educational.under the different trainingconditions, dividede into3groups, blank control(simple MSCs), experimentalgroup (PRF+MSCs) and positive control (BMP-2+MSCs). There are fivesubgroups,they are PRF1ml;PRF2ml;PRF3ml;PRF4mlandPRF5ml. the threegroups ues the way of MTT to detect the cells proliferation rate after24h,48h,72h.By the PNPP to detect the activity of alkaline phosphatas（ALP）after72h..use immunohistochemistry detect collagenⅠand boneY-carboxyglutamicacid (BGP). Through the above methods, detect the cells are osteoblast.resultsBy morphology detection, the treated MSCs from spindle shape intotriangle, cub and polygon shape.MTT detect assay shows the five subgroupsrendered dose response relations,(that means the more doses PRF in subgroup,the more proliferation rate in cells.) contrast with blank group, the result havemeaning of statistics（F=89.548，P=0.000）；contrast with positive control, fromPRF1ml subgroup to PRF3ml subgroup the more doses PRF in subgroup, the more proliferation rate in cells. PRF4ml and PRF5ml subgroups the quantity ofcells are nearly to positive control.there is no meaning of statistics（F=3.364，P=0.142）. ALP activity assay shows the ALP capability of trans-differentedMSCs is higher than MSCS. the five subgroups rendered dose responserelations,(that means the more doses PRF in subgroup, the more activityexpress of ALP in cells.) contrast with blank group, the result have meaning ofstatistics.contrast with positive control, from PRF1ml subgroup to PRF3mlsubgroup the more doses PRF in subgroup, the more activity express of ALP incells. PRF4ml and PRF5ml subgroups the quantity of cells are nearly to positivecontrol.there is no meaning of statistics.IHC shows that blank group, PRF4mlsubgroup and positive group experiment about Type Ⅰcollagen and BGPshows positive.ConclusionsPRF membrane could trans-differentiate rabbit bone marrow mesenchymalcells into obteoblast cells. Those cells who had the peculiatity of osteoblast cells.PRF membrane could provide a new way for Oral implantology.
