Investigation On Preparation Of The Minocycline Hydrochloride Periodontal Film And Parmacodynamics Research

ObjectiveTo select the optimum adjuvants on the minocycline hydrochlorideperiodontal film, determine the prescription and preparation craft of theminocycline hydrochloride periodontal film, research on the SD of ratsperiodontitis treatment effects, provide the scientific basis for further clinicaltrials.Methods(1) Periodontal film preparation: Selecting the appropriate preparationadjuvants of periodontal film at the indexes of vitro dissolution and the drugcontent, then using orthogonal experimental design method, determining thebest prescription and preparation craft of the periodontal film through theinvestigation of the appearance traits and dissolve indicators of periodontal film,monitoring the content of minocycline hydrochloride and controlling theimpurity of the preparation in the minocycline hydrochloride periodontal film byHPLC, so that the quality of the periodontal film makes accords with the filmdemands”chinese pharmacopoeia”2010edition.(2) The survey of periodontal film on periodontitis SD rats: Totally20SDrats were selected and divided into2groups:5as normal control group, therest were made periodontitis model through fischers,method, then thesuccessful periodontitis mode were divided into3groups, control group,Tinidazole periodontal bat drug group, minocycline hydrochloride periodontalfilm drug groug with5in each group. After7days, The changes of body weight and periodontal tissue were detected, simultaneous, detecting GI, PD, TM,ABL, PLI of periodontal indexes in SD rat periodontal tissue, the effect ofminocycline hydrochloride periodontal film on periodontitis SD rat wasobserved.Results(1) Through the vitro dissolution test and orthogonal test,to determine theoptimum adjuvants of periodontal film were PVA(17-88), CMC-Na, the optimalprescription were: minocycline hydrochloride2.00g, PVA(17-88)3.00g,CMC-Na1.50g, Gelatin2mL, distilled water100mL.(2) According to theoptimal prescription of the periodontal film, it can significantly improveperiodontitis SD rat,s body weight, the periodontal tissue was pink, periodontalmembrane was attached normally, trabecular bone was arranged closely, GI,PD, TM, ABL, PBI and other periodontal indexes were improved obviously(P<0.01).ConclusionsThe quality of the minocycline hydrochloride periodontal film makesaccords with the film demands in2010edition chinese pharmacopoeia. Theperiodontal film can significantly improve the periodontitis SD rat,s body weightand the periodontal indexes, which gradually returned to its original level, sominocycline hydrochloride periodontal film have treatment function for theperiodontitis SD rat.
