ObjectiveTo investigate the influence of mast cell to thyroid carcinoma tissue andthe mast cell is promoted by functional stem cell factor and lipopolysaccharidein tumor microenvironment.MethodsTo complete routine culture of mast cell from bone marrow and thyroidcarcinoma SW579cells.To prepare cells suspension and to make nude micetumor-bearing model using SW579cells from thyroid carcinoma. To establishtwo models including SW579cells combined BMMCs cells and intra-tumorinjection BMMCs only and to observe influence to function of mast cell in tumormicroenvironment.To observe influence of mast cell to growth of thyroidcarcinoma tissue through BMMCs which is preprocessing with functional stemcell factor. To simulate tumor microenvironment through NTC-Ms and TI-Msand to observe influence to thyroid carcinoma tissue from mast cell and mixedinoculation of BMMCs and SW579cells preprocessing with NTC-Ms andTI-Ms. To detect functional stem cell factor in NTC-Ms and TI-Ms by cellschemoattractant methods and antibody inhibition experiment. To observeinfluence of mast cell to growth of thyroid carcinoma tissue by mixedinoculation of BMMCs and SW579cells preprocessing with QNZ andresveratrol. To observe influence of mast cell to growth of thyroid carcinoma tissue by mixed inoculation of BMMCs and SW579cells preprocessing withSCF and LPS. To extract total protein from carcinoma above and to detectVEGF and IL-10in tumor by Western blot.ResultsTumor growth is inhibited by mixed inoculation of BMMCs and SW579cells and there is significant difference(P<0.05). Tumor growth is promoted byintra-tumor injection BMMCs only and there is significant difference(P<0.05).The weight of tumor which is preprocessing with SCF is more thanthe other group and here is significant difference(P<0.05). There is significantdifference (P<0.05) in multiple comparison between experimental group andcontrol group.In mixed inoculation model BMMCs has potentiality to promotegrowth of tumor preprocessing with NTC-Ms and TI-Ms. The weight of tumor inexperimental group is higher than control group when concentration ofNTC-Ms is higher than20ug/ml or concentration of TI-Ms is higher than100ug/ml. There is significant difference (P<0.05) in multiple comparisonexcept experimental group four to experimental group five. There is significantdifference (P<0.05) between experimental group and control group. Functionalstem cell factor is contained in NTC-Ms and TI-Ms by cells chemoattractantexperiment. There is significant difference (P<0.05) and effect to promotegrowth thyroid carcinoma tissue is decreased by resveratrol and QNZ throughNTC-Ms preprocessing with MMCs. There is significant difference (P<0.05)and growth of thyroid carcinoma tissuecan be promoted by MMCspreprocessing with SCF and LPS. The high expression of IL-10is stimulatedby SCF,NTC-Ms and TI-Ms by Western blot.ConclusionsPotentiality to promote thyroid carcinoma tissue of mast cell bonemarrow-derived is enhanced by functional SCF and LPS in tumormicroenvironment but concrete mechanism may be clear in further research.
