ObjectiveSalvia Injection preconditioning could reduce liver ischemia-reperfusionreperfusion injury in rats,this study was to explore the mechanism of SalviaInjection ‘s effect on PI3K/Akt signaling pathway and so as to provideexperimental evidence for prevention and treatment for liverischemia-reperfusion injury in clinic.Methods80Clean healthy male Sprague Dawley(SD) rats were randomly dividedinto4groups:sham operation group(sham group), ischemic reperfusiongroup(IR group),Salvia group(SI group),Salvia+LY group(LY group).Reperfusion period in each group was divided into four phases respectively(1,3,6,24h),5rats each phase in each group.70%of hepatic ischemiareperfusion model was made in rats for60minutes ischemia. Serum level ofAlanine aminotransferase (ALT), Aspartate aminotransferase (AST) andLactate dehydrogenase (LDH) were determined,phosphatidyl inositol3-kinase(PI3K), Akt and p-Akt (Ser308) protein expression in liver tissue weremeasured by Western blot method (Western-Blot),hepatic cell apoptosis wasobserved with Terminal deoxynucleotidyl Transferase-mediated dUTP nick endlabeling (TUNEL) method; and morphological changes in rat liver observedunder light microscope. ResultsALT, AST and LDH level in IR,SM and LY group were significantly higherthan those of Sham group (P<0.05).The values of SM group and LY group ateach phase lower compared with those of IR group (P<0.05), and comparedwith LY group, the value of each phase in SM group decreased (P<0.05).In the light of liver morphological changes observed, the most obviousdamage visible in IR group, that of SM group and the LY group lighter than IRgroup, the LY group worse than SM group, while Sham group near to normal.Western-Blot test: PI3K and Akt expression in each group had nosignificant difference (P>0.05); while p-Akt expression in Sham, IR and LYgroup, there was significant differences (P<0.05), The expression in SM groupwas significantly higher than other groups (P <0.05).Apoptosis: The hepatic cells had the typical characteristics of apoptosis inIR group.Apoptosis Index (AI) of SM,LY and sham group were lower than thatof IR group (P<0.05), while that of SM group lower than the LY group(P<0.05),Sham group near to normal, better than the other groups.ConclusionsThe hepatic ischemia-reperfusion caused liver cells significant damage,pretreatment of Salvia Injection could pretect liver function and inhibit liver cellapoptosis by activating PI3K/Akt signaling pathway.
