ObjectiveTo explore the relationship of apoptosis between J774cell and PAsupernatant fluid. To study the expression level of Fas and FAP-1protein duringthe apoptosis.Methods1、J774cells were infected with PA supernatant fluid when PA supernatantfluid∶ J774was1∶15for different lengths of time(3h,6h and9h).2、The J774apoptosis induced by PA supernatant fluid was tested byHoechest33258and Annexin-V-FITC/PI.3、Western blot was used on the quantitative detection of Fas and FAP-1.ResultsHoechst33258fluorochrome staining showed that J774cells in the9hgroup(PA supernatant fluid∶ J774=1∶15) underwent typical apoptosismorphological changes and apoptotic bodies were observed.Hoechst33258fluorochrome staining and Annexin V FITC/PI assay showed that J774cellapoptosis rate is higher with the extension of time. The difference of apoptosisrate of each group compared with the control group are significant (P <0.01);The comparison between the differences are statistically significant (P <0.01).There were no statistical diferences between Hoechst33258fluorochromestaining and Annexin V FITC/PI assay (P>0.05). Western blot showed that the expression level of Fas was enhanced obviously after infecting J774cell for3h.The expression of FAP-1protein was descended.Conclusions1、PAsupernatant fluid can induce apoptosis to J774cell.2、PA supernatant fluid induced apoptosis to J774cell with timedependence.3、The mechanisms of PA supernatant fluid to J774cell apoptosisinvolvedFas and FAP-1protein expression.
