ObjectiveTo explore the effect of HO-1high expression on OS and ERS of themyocardium of type2diabetic rats.MethodsA total of44healthy male SD rats weighting180-200g were used for exper-iment.10rats were randomly chosen as normal control group(NC),and theywere fed with normal feed.The rest34rats that were fedwith high sugar, high fatand high cholesterol diet. After six weeks, these rats were induced to diabetesmellitus by injecting STZ into the caudal vein. Only30of34rats became diabe-tes succesfully, and they were randomly divided into three groups: diabeticgroup (DM), diabetic+heme group(HE) and diabetic+ZnPP group (ZN). Eachgroup contains10rats. In NC rats, cushion fluid was injected into the caudalvein.When the diabetic model was successfully established, Hemin (15mg/kg)and ZnPP(50u mol/kg) were injected into the abdomens of HE and ZN grouprats, respectively, at third day. In NC and DM rats, same amount of normalsaline was injected into the abdomens twice per week. All rats were killed at theninth week. Myocardial oxidative stress level was detected by colorimetricmethod; Myocardial collagen fiberswas detected by masson; The change ofmyocardial ultrastructure was observed under electron microscope; MyocardialHO-1express was measured by immunohistochemistry; the level of myocardialCHOP express were measureed by western btot.Results1. The level of SOD and GSH-PX in HE group are increased as comparedwith DM and ZN groups.and the differences were significant(P<0.05,P<0.01). The level of MDA in HE group are lower than DM and ZN group,and the differe-nces were significant (P<0.01).2. The degree of myocardial fibrosis and theultrastructural damage of myo-cardial mitochondria in group HE are moreserious as compared with DM and ZN group, and the differences were signify-cant (P<0.01).3. The expression level of HO-1in HE group is higher.as comp-ared with DM, NC and ZN group, and the differences were significant (P<0.05,P<0.01,P<0.01).4. The expression level of CHOP in HE group is decreasedthan DM and ZN group, and the differences were significant (P<0.05, P<0.01).But, the expression level of CHOP in HE group is increased as com-pared withgroup NC, and the differences were significant (P<0.05).5. There is a negativecorrelation between HO-1and MDA expression, and the Pearson correlationcoefficient is0.720(P<0.01);There is a positive correla-tion between HO-1andSOD, GSHP-X expression, and the Pearson correlation coefficient are0.799,0.894respectively (P<0.01);There is a negative correlation between HO-1andCHOP expression,and the Pearson correlation coefficient is0.870(P <0.01).There is a negative correlation between CHOP and MDA expression, and thePearson correlation coefficient is0.828(P<0.01).Conclusions1、Heme can induce HO-1express in myocardium myocardium of type2diabetic rats.2、There are oxidative stress and endoplasmic reticulum stress in themyocardium of Type2diabetic rats, and they are influenced each other.3、Incresed HO-1expression can attenue myocardial damage throughenhance the oxidative stress and the endoplasmic reticulum stress of themyocardium of Type2diabetic rats.
