Objective To study the role of phosphatidylinositol-3-kinases (PI3K) and protein kinase B (Akt) in children hemangioma endothelial cells.MethodSpecimens of20proliferating phase hemangioma and20involuting phase were classified by Mulliken’s classification and the expression of proliferating cell nuclear antigen(PCNA).The expression of PI3Kp85(n=20) and p-Akt (n=20) were detected by immunohistochemistry method, and PI3Kp85-mRNA (n=11) and p-Akt-mRNA (n=11) were detected by reverse transcription polymeras chain reaction.Result1.Hemagioma specimens were classified as20proliferating phase hemagioma specimens,20involuting phase hemagioma specimens. In the hemagioma proliferating phase specimens, the expression rate and strength of PI3Kp85and p-Akt were higher than them in the involuting phase specimens(P<0.05, P<0.01).2.In the proliferating phase hemagioma specimens,the integral optical density (IOD) of PI3Kp85and p-Akt expressed were191.23±40.45,78.73±38.49,In the involuting phase hemagioma specimens,the IOD of PI3Kp85and p-Akt expressed were46.63±13.08,64.93±21.22. In the hemagioma proliferating phase specimens, the IOD of PI3Kp85and p-Akt expressed was obviously higher than the IOD in the involuting phase specimens (P<0.01).Meanwhile PI3Kp85and p-Akt have Positive correlation (r=0.553, P<0.05).3.The mRNA relative expression of PI3Kp85and p-Akt were0.0807±0.0136,0.0841±0.0137.In the involuting phase hemagioma specimens, The mRNA relative expression of PI3Kp85and p-Akt were0.0203±0.0098,0.0324±0.0178.In the hema-gioma proliferating phase specimens.The mRNA relative expression of PI3Kp85and p-Akt was obviously higher than it in the involuting phase specimens (P<0.01). Meanwhile the mRNA relative expression PI3Kp85and p-Akt have Positive correlation (r=0.5503,P<0.01).Conclusion:The protein and mRNA of PI3Kp85, p-Akt are expressed in hemangioma vascular endothelial cells.PI3K/Akt signaling pathway may be involved in hemangioma vascular endothelial cells apoptosis.
