Objective:To investigate the expressions of estrogen receptor alpha (ERa) and cyclinDl in human papillary thyroid carcinoma (PTC) tissues.Methods:By means of the technology of Envision immunohistochemical staining, the expressions of ERa and cyclinD1were detected in147tissue samples, all of which were obtained from the thyroid excision specimen of patients with thyroid diseases surgically treated in our department and been pathologically diagnosed. These samples were including48samples of papillary thyroid carcinoma,69cases of benign thyroid disease (28cases of thyroid follicular adenoma,34cases of nodular goiter,6cases of chronic lymphocytic thyroiditis,1case of Hurthle cell tumor) and30cases of normal thyroid gland beside benign thyroid disease tissue.Results:1. The ERa expression can be detected in tissues from normal thyroid gland, benign thyroid disease and PTC, most of them were located within nucleus area. The proportion of ERa positively-expressed tissues in PTC group is25.0%(12/48), which is significantly higher than that in benign thyroid disease group [10.1%(7/69)] as well as that in normal thyroid gland group [6.7%(2/30)](X2=6.595, P<0.05). In samples from PTC group, no statistical significances were found in the difference of ERa expression among patients with different status of sex, age, lymph node metastasis and tumor coat violation(P>0.05).2. The expression of cyclinD1can not be detected in the normal thyroid tissues, while in tissues from benign thyroid disease and PTC, it can be found mainly located in the area of cell nucleus. The proportion of cyclinD1expression in tissues of PTC group is79.2%(38/48), significantly higher than that in the benign thyroid disease[27.5%(19/69)](X2=55.658, P<0.05). Further more, in PTC group this proportion come to be as high as92.9%(26/28) in patients with lymph node metastasis, significantly higher than those without [60.0%(10/12)](X2=5.774, P<0.05). Such difference can also be found in patients with tumor coat violation [90.3%(28/31)] or without [58.8%(10/17)](X2=4.833, P <0.05); but not in patients differ in sex and age (P>0.05).3.The difference between the expression of ERa and cyclinD1in tissues from PTC group is statistically significant.(X2=22.321, P<0.05).Conclusion:1:The ratio of tissues expression of ERa in PTC is significantly higher than that in normal thyroid gland tissues and benign thyroid disease; PTC is probably an estrogen-dependent tumor.2. The protein of cyclinD1are positively expressed in tissues of PTC group while negatively in the normal thyroid tissues. The expression of cyclinD1has relationship with lymph node matastasis and tumor coat violation in PTC patients, which may suggest a poor prognosis of this disease.3. The difference of the expression between ERa and cyclinDl in PTC has a statistical significance.4. Detections of the tissue expression of cyclinD1and ERa are helpful to further clarify the pathogenesis and predict the recurrence risk of PTC. The treatment with estrogen inhibitor and anti-cyclinD1may be a new therapeutic approach to PTC.
