Objective:The purpose of this study was to detect Snail and NF-κB in epithelial ovarian carcinoma, analyse the relationship between the expression of both and clinical pathological feature of epithelial ovarian cancer. The correlation of Snail and NF-κB was also discussed. Through this research, we aim to provide pathological and molecular biological basis for the progression, metastasis of epithelial ovarian cancer, so that new detecting and treating methods of ovarian cancer was approaching.Methods:59ovarian neoplasm wax blocks was collected from pathology department of Yang Zhou NO.1hospital from2005to2010and BaoYing people’s hospital from2007to2010, meanwhile,18benign tumor specimens and10normal ovarian specimens were collected as control group. The patients were received operation as soon as ovarian cancer was diagnosed without any chemotherapy or radiotherapy before surgery. All specimens were fixed by10%formaldehyde. Through paraffin embedding, the specimens were made into tissue sections. After HE staining and microscopy, pathological diagnosis and sampling parts were ascertained. The expression of Snail and NF-κB were detect by immunohistochemical. Within the results, we employed spss16.0to analyze the relationship between the expression of Snail, NF-κB and clinicopathologic parameters, such as histological type, histological grade, FIGO clinical stage and lymph node metastasis.Results:1. Positive snail expression in normal ovarian specimen, benign epithelial ovarian tumors and malignant epithelial ovarian tumor were0.00%(0/10),16.67%(3/18),76.27%(45/59) respectively. According to FIGO clinical stage, stage Ⅰ-Ⅱ was41.18%(7/17), stageⅢ-Ⅳ was90.47%(38/42). Positive snail expression in ovarian carcinoma was significant between benign and normal(P<0.005), meanwhile, the expression of Snail in stageⅢ-Ⅳ was higher than that in stage Ⅰ-Ⅱ, it’s statistically significant(P<0.005). However, there was no significant between benign and normal ovarian benign tumor (P>0.05).The relationship between the expression of Snail, and clinicopathologic parameters: Statistical analyse showed that positive expression of Snail in epithelial ovarian carcinoma was closely correlated with clinicopathologic parameters. The positive expression of Snail in serous ovarian carcinoma was higher than mucinous (P<0.005). It is significant between high and low histological grade (P<0.05) and it was also the same between FIGO clinical stageⅢ-Ⅳ and stage Ⅰ-Ⅱ (P<0.005). The higher of CA125(≥600U/L), the stronger Snail expression was (P<0.001). It has identical situation in diameter of tumors (P<0.005). In case of lymph node metastasis in epithelial ovarian carcinoma, it was higher Snail expression than those no lymph node metastasis (P<0.005). Whereas, there was no significant in age of patients2. NF-κB expression in normal ovarian specimen, benign epithelial ovarian tumors and malignant epithelial ovarian tumor were20.00%(2/10),27.78%(5/18),57.63%(34/59) respectively. Positive NF-κB expression in ovarian carcinoma was significant from benign and normal (P<0.05). However, there was no significant between benign and normal ovarian specimen (P>0.05).The relationship between the expression of NF-κB, and clinicopathologic parameters:According to this research, positive expression of NF-κB in epithelial ovarian carcinoma was significant in histological grade (P<0.05), size of tumor diameter (P<0.05). Just like the expression of Snail, the higher of CA125, the stronger NF-κB expression was (P<0.005). As for lymph node metastasis, it was significant between positive and negative (P<0.005). There was no statistically significant in histological type, clinical stage.3. The correlation between Snail and NF-κB:the positive expression rate of NF-κB in both groups with and without Snail expression was56.60%(30/53),28.57%(4/14), the difference between two groups was significant (P<0.05), and was significantly positive correlation with each other(r=0.312).Conclusion:1.There was up-regulated expression of Snail protein in epithelial ovarian carcinoma. The higher stage with stronger expression.2.Snail facilitates the programme of ovarian cancer metastasis. Positive expression of Snail in metastasis is associate with poor prognosis. There was clinical meaning in detecting Snail in facing borderline ovarian tumors.3.Snail was a potential target in the inhibition of ovarian cancer metastasis.4. The expression of NF-κB in ovarian cancer was strong. NF-κB may play a role in ovarian cancer progression. Besides, NF-κB expression was correlated with Snail. It may play cooperation role with Snail in the metastasis.
