In this paper, the antitumor and immunopotentiation activities of Fomitopsis pinicola’s fermentation products and monomericcompound (3-acetoxylanosta-8,24-dien-21-oic acid) were studied,and Fermentation process research of Fomitopsis pinicola were studied,and on that basis, an antitumor health product (Fomitopsis pinicola Antitumor Tablet) was developed.In this paper, Fomtopsis pinicola as a spawn with standard fermentation medium was Screened, The results show that the optimum standard fermentation medium:potato200g/L, glucose20g/L, KH2PO41g/L, MgSO41.5g/L, vitamin B10.4g/L; the optimum fermentation conditions of fermentater:the best fermentation period is56-64h,the weight of dry mycelium6.8901g/L, the content of Exopolysc-charides (EPS)0.2965g/L.In this paper, the antitumor activities of fermentation product and monomericcompound (3-acetoxylanosta-8,24-dien-21-oic acid) which was isolated from chloroform extract were studied, the anti-tumor activities of these extracts and the monomer compound were studied in vivo, the compounds of monomer compounds was researched by cell apoptosis experiment. Effects of extracts on immune organ in H22mice were detected with measuring the thymus index and spleen index. The results showed that the fermentation product and monomericcompound had a higher inhibition rate. The inhibition rate of monomericcompound was63.09%a on the dose of5mg/kg/d. The thymus index of chloroform was significantly higher than the CTX groups(P<0.05), closing to the normal group(P>0.05). And the inhibition rate of fermentation product was56.76%a on the dose of100mg/kg/d (its content is equivalent to monomer compounds of0.3mg/kg/d). The thymus index of chloroform was significantly higher than the CTX groups(P <0.01), closing to the normal group(P>0.05). The apoptosis rate of cancer cell of the monomericcompound is only23.2%in the experiment, Close to20.9%of the negative control group, promote cell apoptosis effect is not obvious. Based on the experimental results in vivo and in vitro, compound A was determined as one of the compositions of antitumor of Fomtopsis pinicola. The antitumor effect may realize by increasing the body’s immune activity, but its mechanism remains to be studied in the future.The immunopotentiation activities of dry mycelia of Fomitopsis pinicola was researched for the model with mice for the first time. The Organ immune index, devour rate of rat’s peritoneal macrophage englobing chicken red blood cell, serum hemolytic level, the percent conversion of ConA induce the transformation of spleen lymphocytes of mice and the clearance rate of carbon particles. The result of the experiment of devour rate of rat’s peritoneal macrophage englobing chicken red blood cell shows that the devour rate of the experimental group reached72.8%,which was Obviously higher than the result with51%of the negative group (P<0.01).In the experiment of serum hemolysin,the serum hemolytic level of the experimental group reached54.678,which was Obviously higher than the result with37.532of the negative group (P<0.01).And in the experiment of ConA induce the transformation of spleen lymphocytes,the percent conversion shows that the devour rate of the experimental group reached55.333%,which was Obviously higher than the result with35.501%of the negative group (P<0.01). The result of experiment of the carbon clearance test of mice shows that after the action of the drug, the index of carbon clearance increased dramatically.From above results, the experimental group is much better than the control group on survival condition, devour rate, serum hemolytic level,lymphocytetransformationrate and the clearance rate of carbon particles. The experimental results prove that the dry mycelia of Fomitopsis pinicola can Improve the immunity activity of the mice. But its mechanism remains to be studied in the future.In this paper, an antitumor health product—Fomitopsis pinicola Antitumor Tablet was developed by the method of compressing dry granulation. And the dose of the Fomitopsis pinicola Antitumor Tablet was determined of Fomitopsis pinicola’s fermentation products10g, lactose20g, MCC20g, magnesium stearate0.5g. The content of compound (3-acetoxylanosta-8,24-dien-21-oic acid) in fermentation product of Fomitopsis pinicola was measured in the experiment, the result showed that the content of the compound in the mycelium reached0.314%.The toxicology of fermentation product of Fomitopsis pinicola was studied preliminary in the experiment, the result showed that the mice were all alive in the experimental determination of LD50,and didn’t have any sign of poisoning. So the LD50was not measured. In the experiment of measuring the maximum tolerance of mice, when the dose of medication reached to8000mg/Kg,the mice were still alive. So we can come to a conclusion that the fermentation product of Fomitopsis pinicola did not have virulence, and the stability, usage and dosage, preserving instruction of Fomitopsis pinicola Antitumor Tablet was studied preliminarily, the stability of the tablet was very good, and it could be preserved and eaten easily, so the Fomitopsis pinicola Antitumor Tablet was a new-style antitumor health product which has a large-scale potential of production and development.
