To study the effect of HYSA on the process of experimental hepatic fibrosis in rats and the mechanism.1. Liver fibrosis model was made in rats by perfusion with carbon tetrachloride. The rats were divided into five groups:the normal group、the model group、the safflower group、the HYSA group and the Colchicine group. Appropriate drugs were intraperitoneal injected from the second week of modeling except the normal group:1ml NS/kg、500mg Safflower/kg、10mg HYSA/kg、0.1mg Colchicine/kg. All the drugs were given once a day,five times every week. The treatment lasted8weeks for all the groups. Serum ALT、AST and ALB were detected. Hematoxylin and eosin staining of liver tissue was performed for histopathological evaluation of liver fibrosis. Hoechest33342fluorescent staining of liver tissue was performed to observe appearance of cell nucleus.2. Serum HA、LN、PCⅢ、CⅣ were detected by radioimmunity. HYP contents in liver tissue were detected by alkaline hydrolysis. Masson’s trichrome staining of liver tissue was performed for histopathological evaluation of liver fibrosis.3. SOD and MDA contents in liver tissue were detected.The expression of a-SMA、TIMP-1and TGF-β1in liver tissue was detected by immunohistochemistry. The results showed that:1. Compared with the model group, the levels of serum ALT、AST were significantly decreased, while the level of serum ALB was significantly increased, Apoptosis of liver cells were less and the level of fibrosis were abatement in the safflower group、the HYSA group and the Colchicine group.2. Compared with the model group, serum HA、LN、PCIII、CIV, areas of fibre and HYP contents in liver tissue were significantly decreased in the safflower group、the HYSA group and the Colchicine group.3. Compared with the model group, MDA contents in liver tissue were significantly decreased, SOD contents in liver tissue were significantly increased in the safflower group、the HYSA group and the Colchicine group. The expression intensity of a-SMA、TIMP-1and TGF-β1proteins were significantly lower in the safflower group、the HYSA group and the Colchicine group. It may be concluded that:HYSA can prevent carbon tetrachloride-induced liver fibrosis in rats. The effect of HYSA is better than safflower. This may be associated with anti-lipid peroxidation and abatement the expression intensity of a-SMA、TIMP-1and TGF-β1proteins.
