Objective:To investigate the Sodium paeonol on experimental immunologic Conjunctivitis of treatment,and to explore its mechanism.Methods:we use chicken ovalbumin (OVA) immune Balb/c mice to establish animal models of immunologic Conjunctivitis; It is divided into normal control group group. model group (PBS eye drops)、1%sodium paeonol group (1%Sodium paeonol eye drops),2%sodium paeonol group (2%sodium paeonol eye drops)、 sodium cromoglicate group, olopatadine group of six groups. After the success in modeling Medication them five days, we observed score of the eye clinical symptoms when after the last treatment,then abdominal venous blood、killed, and get conjunctiva, we use toluidine blue staining To investigate conjunctiva mast cell degranulation in the case, by enzyme-linked immunosorbent assay (ELISA) to detect the experimental animals serum OVA-sIgE, IFN-y and IL-4levels.Results:1.Comparison of eye irritation.The symptom score of model group was significantly higher than normal control group(P<0.01),compared with model group the symptom score were lower in all of the treatment groups(P<0.05or P<0.01), there was no significant effect between1%and2%sodium paeonol group(P>0.05).2.Comparison of conjunctival mast cells. Compared with normal control group the population of conjuctival mast cells was obviously increased in both model group and all of the treatment groups(P<0.01),there was no significant difference between the treatment groups(P>0.05),comared with model group the degranulation of conjunctival mast cells was obviously lower in both normal control group and all of the treatment groups (P<0.01).3. Comparison of the serum level of OVA-sIgE.The serum level of OVA-sIgE of model group was significantly higher than normal control group(P <0.01), compared with model group the level of OVA-sIgE was lower in1%sodium paeonol group,2%sodium paeonol group and olopatadine group (P<0.05or P <0.01),there was no significant difference between1%and2%sodium paeonol group(P>0.05). 4.Comparison of the serum level of IFN-γ. The serum level of IFN-y of model group was obvioulsy lower than normal control group(P<0.01), compared with model group the level of IFN-y was higher in all of the treatment groups (P <0.05or P<0.01).5.Comparison of the serum level of IL-4.The serum level of IFN-y of model group was obvioulsy higher than normal control group(P<0.01), compared with model group the level of IFN-y was lower in all of the treatment groups (P <0.01).6.Correlation analysis of the serum level of OVA-sIgE、IFN-y and IL-4.There was a positive correlation between the level of OVA-sIgE and IL-4,both of the level of OVA-sIgE and IL-4were negatively correlated with thelevel of IFN-y. Sodium paeonol could increase the level of IFN-y and at the same time reduced the level of OVA-sIgE and IL-4.Conclusion:1.Sodium paeonol have a therapeutic effect on experimental immunologic Conjunctivitis.2.Sodium paeonol could inhibit the activation of mast cell degranulation in conjunctival of experimental immunologic Conjunctivitis, and the innibition might be concentration-dependent,2%sodium paeonol have the same inhibit effect as sodium cromoglicate and olopatadine.3.Sodium paeonol could reduce the serum level of OVA-sIgE of experimental immunologic Conjunctivitis.4.Sodium paeonol could reduce the serum level of IL-4and at the same time increased the level of IFN-y,2%sodium paeonol are more effective on IL-4and IFN-y than1%sodium paeonol.
