The Study Of Identification And Metabolites Of Endophytic Fungi1621Strain Of Coptis Deltoidea C.Y.Cheng Et Hsiao(Ranunculaceae)

Endophyte commonly found in plants, many of which endophyte could produce the same and similar metabolites, and many of them are the new substance never found.So, using the endophytic to select bioactive components or leading compounds is an important way to select natural medicine. Studying team separated and selected some strains which has antibacterial activity before.This study select1621strain which has stronger antibacterial activity to study the fermentation condition.and identify it.Using the method combined with morphology and molecular biology to identified the1621strain and the result is that it is a new strain of Aspergillus fumigatus. named CD1621. Study on the fermentation condition of the CD1621strain and drawing the growth curve and fermentation broth pH change curve and determining the most suitable C/N rate for growth is15:1. Through the orthogonal test selection of carbon source、nitrogen source and initial pH value find that the effection from big to small is carbon source、nitrogen source and initial pH value in turn.The best composition of culture medium is soluble starch(3%),(NH4)2SO4(0.2%), the initial pH value is7.Separating6kinds of metabolites from fermentation broth by using macroporous resin, chromatographic silica and preparative liquid and other isolation and purification methods.Determining the structure of6kinds of metabolites by using nuclear magnetism, mass spectrum and IR. The other compounds Ⅰ～Ⅵ are benzoic acid、 palmitic acid、Monomethylsulochrin、Pseurotin A and2-methyl-5-methoxy-6-hydroxy-2,3-epoxide-p-benzoquinoe、5-methyl-3-methoxy-2-hydroxy-2-cyclopentene-1,4-dione.Compound Ⅵ is new which isolated and got from the nature. Determinating the data of nuclear magnetism、IR and optical rotation and improving the structure information of compound V.The flat filter paper method for compound Ⅰ, Ⅲ, Ⅳ, Ⅴ and Ⅵ on Escherichia coli and Staphylococcus aureus were.bacteriostatic activity detection, the results shows that benzoic acid、Monomethylsulochrin and compound Ⅵ have bacteriostatic action against escherichia coli and staphylococcus aureus. Pseurotin A has bacteriostatic action against escherichia coli and compound V has bacteriostatic action against staphylococcus aureus.In summary, both from the morphological analysis and molecular biology, completed the optimization of the endophytic fungi strains1621, identification, fermentation conditions and medium as well as its metabolites separation, identification and activity screening,access to an antibacterial activity of new compounds,laid a foundation for in-depth study of the strain1621.
