Objective:To investigate the pathological changes of vitiligo guinea pig model which induced by5%hydrogen dioxide,and study the influence of pimecrolimus on expression of HMB45,TNF-α and IL-4in lesion of vitiligo guinea pig model.Then get the underlying mechanism of how pimecrolimus induces repigmentation in vitiligo.Methods:This study is divided in two parts.The first part is the establishment of animal of vitiligo. Vitiligo guinea pig model was established by5%hydrogen dioxide,we observed the change of skin melanin content.The second part is investigating the curative effect of pimecrolimus on experimental vitiligo in guinea pigs.The guinea pig models were treated with1%pimecrolimus.Immunohistochemical studies were performed to study the cytokines in1%pimecrolimus treated group,vitiligo model group and normal control group.Results:1.The result showed that under macroscopic observation,the depigmentation of the skin between vitiligo model group and normal control grouphad no significant difference.In the staining method for melanin sections,melanin reduced in the spinous layer and the hair follicles of model group.Compared with normal control group,model group has significant diference(p<0.01).2.The expression of HMB45in lesion of vitiligo model group was lower than normal control group.The expression of HMB45in lesion of pimecrolimus treated group was higher than the vitiligo model group(P<0.01).The expression of TNF-aand IL-4in vitiligo model group was significantly higher than the normal control group(p<0.01).The expression of TNF-α and IL-4in pimecrolimus treated group was significantly lower than the vitiligo model group(P<0.01).Conclusion:1.The guinea pig is elegible for establishing vitiligo animal model by being decolorized with hydrogen dioxide.2. The pimecrolimus has good effects on vitiligo guinea pig model. Pimecrolimus could promote the synthesis of melanin which may relate with inhibit the expression of TNF-α、IL-4and promote the expression of HMB45in lesion of vitiligo model.
